柴胡疏肝散对抑郁症和心绞痛“异病同治”的网络药理学分析

Network pharmacological analysis of Chaihu Shugan powder in treating depressive disorder and angina pectoris with concept of“treating different diseases by same method”

目的:借助网络药理学方法阐释柴胡疏肝散治疗抑郁症和心绞痛的“异病同治”机制。方法:应用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)筛选柴胡疏肝散组成药味的成分并预测成分靶点。应用DrugBank、OMIM、GeneCards、CTD数据库查询抑郁症和心绞痛靶点。应用TBtools软件取成分和疾病交集靶点,导入STRING数据库构建蛋白互作(Protein-protein Interaction,PPI)网络,导入David数据库进行富集分析。结果:查询得到柴胡疏肝散中符合筛选标准的化合物共143个,成分涉及靶点141个,抑郁症靶点3296个,心绞痛靶点178个,最终得到交集靶点44个,其中自由度大于均值的关键节点共23个,如TP53、TNF、IL-6、MAPK1、JUN等。David数据库富集得到衰老、凋亡过程的负调节、细胞对脂多糖的反应、脂多糖介导的信号通路、一氧化氮生物合成过程的正调控等265条生物过程及TNF、HIF-1、NOD样受体、PI3K-Akt、T细胞受体信号通路等95条通路信息。结论:柴胡疏肝散对抑郁症和心绞痛“异病同治”的机制主要涉及炎症反应、氧化应激、免疫调控等通路,为进一步对柴胡疏肝散“异病同治”抑郁症和心绞痛的实验验证提供科学依据。

Objective:To explore the mechanism of Chaihu Shugan powder(柴胡疏肝散,CHSGP)treating depressive disorder and angina pectoris with concept of“treating different diseases by same method”by network pharmacology method.Methods:TCMSP was used to screen the compounds and targets of CHSGP.Drugbank,OMIM,GeneCards and CTD were used to find targets of depressive disorder and angina pectoris.Compounds and diseases intersection targets were obtained by TBtools software.Protein-protein interaction was constructed by STRING database and enrichment analysis was conducted by David database.Result:A total of 143 kinds of compounds,141 compound targets,3296 depressive disorder targets,and 178 angina pectoris targets were collected from CHSGP.Finally,44 common targets were obtained,including 23 key targets with degree greater than the mean,such as TP53,TNF,IL-6,MAPK1,JUN,etc.265 biological processes included aging,negative regulation of apoptotic process,cellular response to lipopolysaccharide,lipopolysaccharide-mediated signaling pathway,positive regulation of nitric oxide biosynthetic process and 95 pathways included TNF,HIF-1,NOD-like receptor,PI3K-Akt,T cell receptor signaling pathway were obtained by enrichment analysis.Conclusion:The mechanism of CHSGP treating depressive disorder and angina pectoris with concept of“treating different diseases by same method”is related to inflammatory response,oxidative stress,immune regulation and other pathways,which provids scientific basis for further experimental verification.
