摘要
目的:观察星状神经节阻滞(SGB)对体外循环(CPB)下大鼠海马神经元凋亡的影响。方法:将40只SD大鼠随机分为4组,即假手术组(S组)、SGB组、CPB组、SGB+CPB组,于CPB后2 h处死大鼠,取大脑组织和海马组织。苏木精—伊红(HE)染色检查海马神经元损伤情况;TUNEL法检测海马组织细胞凋亡;免疫组化法检测Bax蛋白表达;western blotting检测细胞色素c(Cyt-c)和Caspase-3蛋白表达。结果:SGB组可缓解由CPB引起大鼠海马神经元损伤。与S组比较,CPB组和SGB+CPB组海马神经元凋亡指数及Cyt-c、Bax和Caspase-3蛋白表达量升高(均P<0.05);与CPB组比较,SGB+CPB组海马神经元凋亡指数及Cyt-c、Bax和Caspase-3蛋白表达量降低(均P<0.05)。结论:SGB可减轻由CPB引起的大鼠海马神经元凋亡,其机制可能与调节线粒体凋亡途径有关。
Objective:To study the effect of stellate ganglion block(SGB)on neuronal apoptosis of rats after cardiopulmonary bypass(CPB).Methods:Forty SD rats were randomly divided into 4 groups:sham-operatedgroup(S group),SGB group,CPB group and SGB+CPB group.Rats were sacrificed 2 hours after CPB.Brain tissues and hippocampal tissues were collected.Hippocampal neuron damage was examined by HE staining.The apoptotic rate was assessed by TUNEL staining.The expression of Bax was detected by immunohistochemistry,and the expression of cytochrome C(Cyt-c)and Caspase-3 was detected by western blotting.Results:SGB could alleviate hippocampal neuron injury caused by CPB in rats.Compared with sham-operated group,the apoptotic rate(apoptosis index)and the expression levels of Cyt-c,Bax and Caspase-3 were increased in CPB group and SGB+CPB group(P<0.05).Compared with CPB group,the apoptosis index and the expressions of Cyt-c,Bax and Caspase-3 were decreased in SGB+CPB group(P<0.05).Conclusion:SGB can alleviate the apoptosis of rat hippocampal neurons caused by CPB,and the mechanism may be related to the regulation of the mitochondrial apoptosis pathway.
