藏药亚吉玛(长梗金腰)醋酸乙酯部位的化学成分研究 被引量:2

Isolation and identification of chemical constituents from Tibetan medicine “Ya-Ji-Ma”(Chrysosplenium axillare)

摘要 目的研究藏药亚吉玛(长梗金腰Chrysosplenium axillare)醋酸乙酯部位的化学成分。方法采用HP-20大孔吸附树脂、Sephadex LH-20凝胶等柱色谱以及制备高效液相技术进行化学成分分离,采用NMR和HR-ESI-MS等光谱方法进行化合物结构鉴定。结果分离得到17个高度氧化的黄酮醇衍生物及4个三萜类化合物,其结构分别鉴定为5,6,4’-三羟基-3,7,3’-三甲氧基黄酮-6-O-β-D-葡萄糖苷(1)、5,6,3’-三羟基-3,7,4’-三甲氧基黄酮-6-O-β-D-葡萄糖苷(2)、chrysosplenoside H(3)、5-O-demethylapulein (4)、5,3’-二羟基-3,6,7,4’-四甲氧基黄酮(5)、5,2’-二羟基-3,7,4’,5’-四甲氧基黄酮(6)、chrysosplenol C (7)、5,2’,5’-三羟基-3,7,4’-三甲氧基黄酮(8)、axillarin (9)、brickellin (10)、槲皮素(11)、芦丁(12)、isorientin (13)、chrysosplenosideE (14)、槲皮素3-O-β-D-葡萄糖苷(15)、5,6,3’,4’-四羟基-3,7-二甲氧基黄酮-6-O-β-D-葡萄糖苷(16)、tomentin(17)、β-peltobo1toykinolic acid(18)、3α,16α-二羟基-4,4,9,14-四甲基-(9β,10α)-19-去甲孕酮-5-烯-11,20-二酮(19)、3α,16α,20,25-四羟基-(10α)-葫芦素-5-烯-11,22-二酮(20)、6α,23α-环氧-3β,20β-二羟基-10αH,23βH-葫芦素-5,24-二烯-11-酮(21)。结论化合物2、4~6、8、9、19~21为首次从金腰属植物中分离得到,其余化合物均为首次从长梗金腰中分离得到。 Objective To isolate and identify the chemical constituents from the ethyl acetate fraction of the Tibetan medicine Chrysosplenium axillare. Methods HP-20 macroporous adsorption resin, Sephadex LH-20 gel column chromatography and preparative high performance liquid chromatography were used to separate the chemical constituents. The structures of all isolates were elucidated by spectroscopic methods, including NMR and HR-ESI-MS. Results Seventeen highly oxidized flavonol derivatives and four triterpenoids were isolated and their structures were identified as 5,6,4′-trihydroxy-3,7,3′-trimethoxyflavone-6-O-β-D-glucoside(1), 5,6,3′-trihydroxy-3,7,4′-trimethoxyflavone-6-O-β-D-glucoside(2), chrysospenoside H(3),5-O-demethyllapulein(4), 5,3′dihydroxy-3,6,7,4′-tetramethoxyflavone(5), 5,2′-dihydroxy-3,7,4′,5′-tetramethoxyflavone(6),chrysosplenol C(7), 5,2′,5′-trihydroxy-3,7,4′-trimethoxyflavone(8), axillarin(9), brickellin(10), quercetin(11), rutin(12), isorientin(13), chrysosplenoside E(14), quercetin-3-O-β-D-glucoside(15), 5,6,3′,4′-tetrahydroxy-3,7-dimethoxyflavone-6-O-β-D-glucoside(16), tomentin(17), β-peltoboykinolic acid(18), 3α,16α-dihydroxy-4,4,9,14-tetramethyl-(9β,10α)-19-norpregn-5-ene-11,20-dione(19), 3α,16α,20,25-tetrahydroxy-(10α)-cucurbit-5-ene-11,22-dione(20), 16α,23α-epoxy-3β,20β-dihydroxy-10αH,23βH-cucurbit-5,24-dien-11-one(21). Conclusion It is the first report for the compounds 2, 4—6, 8, 9, 19—21 identified in the genus Chrysosplenium. The other compounds were isolated from C. axillare for the first time.
