摘要
目的研究木荚红豆叶Ormosia xylocarpa 70%乙醇提取物的醋酸乙酯萃取物化学成分及其对肝癌HepG2的细胞毒性作用。方法利用多种色谱学方法和波谱学技术分离鉴定化合物,采用CCK-8比色法测定化合物的细胞毒活性。结果从木荚红豆叶70%乙醇提取物的醋酸乙酯萃取物中分离鉴定8个化合物,分别为3’,4"-二羟基-3,5,4’,3",5"-五甲氧基-7,9’;7’,9-双环氧-4,8"-氧-8,8’-倍半木脂素-丙醇(1)、染料木素(2)、槲皮素(3)、羽扇豆酮(4)、β-香树脂醇(5)、蒲公英赛醇(6)、mundulin(7)和齐墩果酸(8)。化合物7对人肝癌HepG2细胞有细胞毒性作用,半数抑制浓度(median inhibition concentration,IC50)值为86.34μmol/L。结论所有化合物均首次从该植物中分离得到,其中化合物1为新的四氢呋喃类木脂素,命名为木荚红豆素;化合物5~8为首次从红豆属中分离得到,化合物7具有一定的细胞毒活性。
Objective To study the chemical constituents from the ethyl acetate fraction of the 70% ethanol extract from the leaves of Ormosia xylocarpa and their cytotoxic activities. Methods The compounds were separated by a variety of chromatographic methods and identified by spectroscopy techniques. The cytotoxic activities were tested by CCK-8 assay. Results Eight compounds were isolated from the ethyl acetate fraction of the 70% ethanol extract from the leaves of O. xylocarpa, which were identified as 3’,4 "-dihydroxy-3,5,4’,3",5 "-pentamethoxy-7,9’;7’,9-diepoxy-4,8"-oxy-8,8’-sesquineo-lignan-propanol(1), genistein(2), quercetin(3), lupinone(4), β-anulanol(5), epitaraxerol(6), mundulin(7), and oleanolic acid(8). Compound 7 showed moderate inhibition on HepG2 cells, with a IC50 value of 86.34 μmol/L. Conclusion All compounds are obtained from this plant for the first time, among them, compound 1 is a new lignan, namely xylocarpalignan;Compounds 5—8 are obtained from the Ormosia for the first time.Compound 7 shows moderate inhibition on HepG2 cells.
