摘要
目的 构建大鼠实验动物模型,评价BMP2和TGF-β1调控iRoot BP Plus直接盖髓后修复性牙本质形成过程的影响。方法 iRoot BP Plus为对照组、iRoot BP Plus+BMP2组、iRoot BP Plus+TGF-β1组为实验组进行盖髓,在术后14、28、42 d取材,提取RNA,进行荧光定量RT-PCR法观察SIBLING家族在大鼠牙体组织中的表达。结果 术后14 d,BMP2和TGF-β1能上调IBSP、DMP1、SPP1和MEPE的表达水平,TGF-β1作用更显著。术后28 d时TGF-β1能促进DMP1、IBSP及MEPE的表达,BMP2作用不显著。术后42 d时TGF-β1仅能显著促进IBSP的表达。结论 TGF-β1和BMP2可能通过上调SIBLING家族中DMP1、IBSP及MEPE的表达,促进iRoot BP Plus在直接盖髓后修复性牙本质的形成。
Objective To evaluate the effect of BMP2 and TGF-β1 on the dentin repair function of iRoot BP Plus after direct pulp capping. Methods The iRoot BP Plus+BMP2 and the iRoot BP Plus+TGF-β1 group were the experimental group and iRoot BP Plus group served as control. RNA was extracted on 14 days, 28 days and 42 days after pulp capping.The expression of SIBLING family in dental tissue of rat was observed by real-time RT-PCR. Results The expression levels of IBSP, DMP1, SPP1, and MEPE in the dental tissues were significantly up-regulated by BMP2 and TGF-β1 after 14 days. The up-regulation of TGF-β1 was more significant. At 28 days postoperatively, TGF-β1 could significantly promote the expression of DMP1, IBSP and MEPE. TGF-β1 promoted the expression of IBSP at 42 days after surgery.Conclusions TGF-β1 and BMP2 may increase the expression levels of DMP1, IBSP and MEPE in the SIBLING family,promoting the formation of repairing dentin after iRoot BP Plus capping.
