摘要
随着仪器分析技术的进步和高校科研条件的改善,大型仪器已经在高校实验室中得到了进一步的普及。基质辅助激光解吸离子化质谱成像(Matrix assisted laser desorption ionization mass spectrometry imaging, MALDI MSI)技术作为新型的分子原位成像技术,已经被广泛应用于化学、医学、刑侦学等领域中不同类型样本的分析与研究。MALDI MSI虽然应用广泛,但是实验过程繁琐耗时,难以在高校中开展实验教学。为此设计了以磷脂化合物标准品为样本的质谱成像实验,通过简化实验步骤,将复杂的MALDI MSI实验补充到本科生实验教学中,有助于学生了解MALDI MSI的实验原理,掌握MALDI MSI的样本前处理过程和实验数据的采集与处理方法,并使学生了解目前分析领域的新型研究方法和手段,拓宽学生的科研视野。
In recent decades, with the progress of instrumental technology and the improvement of universities’ scientific research conditions, large-scale precision instruments have been further popularized in universities. As a new in-situ molecular imaging technique, MALDI MSI has been widely used in many fields, including chemistry, medicine and forensic sciences. Despite its many advantages, the experimental process of MALDI MSI is too complicated and time-consuming to carry out in the universities’ experimental teaching. This study aims at simplifying the experimental steps by using the phosphatide standards as the imaging sample and ensure that the experiment can be used for undergraduate teaching. The MALDI MSI experiment can help students understand the principle of MALDI MSI, master MALDI MSI sample pretreatment process and acquire proficiency in processing experiment data, and broaden scientific vision.
