miRNA-125b、miRNA-34a在不同分型鼻息肉中的表达与相关性分析 被引量:1

The expression and correlation analysis of miRNA-125b and miRNA-34a in different types of nasal polyps

摘要 目的探讨miRNA-125b、miRNA-34a水平在不同分型鼻息肉中的表达与相关性。方法选取2020年3月至2021年3月收治的初发鼻息肉及复发鼻息肉患者51例,正常对照20例。鼻息肉患者收集患者切除的鼻息肉组织,对照组收集鼻黏膜组织作为试验材料,共收集标本71份;采用全血细胞自动分析仪进行(嗜酸性粒细胞,EOS)水平检测,当EOS比例>5.0%时记录为阳性(+);采用荧光定量PCR检测miRN-125b和miRN-34a的表达在鼻黏膜组织及息肉组织中的表达,通过列联系数法分析miRNA-125b、miRNA-34a水平与不同分型鼻息肉相关性。结果正常组织、初发鼻息肉组织、复发鼻息肉组织中EOS的阳性率分别为10.00%、70.37%、83.33%,不同类型鼻息肉组织与正常鼻黏膜组织中EOS阳性率比较,差异有统计学意义(P<0.05);初发组、复发组标本组织中miRNA-125b平均水平约为正常标本组织的4倍,miRNA-34a的平均表达水平分别接近正常标本组织2倍,各组间miRNA-125b水平存在显著差异(P<0.05),miRNA-34a的水平也存在显著差异(P<0.05);EOS与miRNA-125b的相关系数为0.375,与miRNA-34a的相关系数为0.283。结论miRNA-125b、miRNA-34a水平与EOS水平及鼻息肉的发病存在密切相关性,此相关性对鼻息肉靶向治疗及预后具有重要意义。 Objective To investigate the expression levels and correlation of miRNA-125b and miRNA-34a in different types of nasal polyps.Methods A total of 51 patients with first-onset nasal polyps and recurrent nasal polyps were enrolled in the study,and 20 healthy subjects were enrolled as control group.The,nasal mucosa tissues and nasal polyp tissues were collected respectively from control group and case group.and a total of 71 specimens were collected.The levels of eosinophil,EOS were detected by whole blood cell autoanalyzer.When the EOS ratio was greater than 5.0%,it was recorded as positive(+),and fluorescence quantitative PCR was used to detect the expression of miRN-125b and miRN-34a in nasal mucosa tissue and polyp tissue.Moreover the correlation between miRNA-125b,miRNA-34a levels and different types of nasal polyps was analyzed by column connection number method.Results The positive rates of EOS in normal tissue,initial nasal polyp tissue,and recurrent nasal polyp tissue were 10.00%,70.37%,83.33%,respectively.There were significant differences in the EOS positive rates between different types of nasal polyp tissue and normal nasal mucosa tissue(P<0.05).The average levels of miRNA-125b in initial group and relapse group were 4 times as high as those in control group,and the average expression levels of miRNA-34a were 2 times as high as those in control group.There were significant differences in the expression levels of miRNA-125b and miRNA-34a among groups(P<0.05).In addition the correlation coefficient between EOS and miRNA-125b was 0.375,which was higher than that(0.283)of miRNA-34a.Conclusion The expression levels of miRNA-125b and miRNA-34a are closely related to those of EOS,which are closely correlated with the pathogenesis of nasal polyps,which is of great significance for targeted therapy and prognosis of nasal polyps.
