基于数据挖掘的毒性中药配伍减毒规律分析

Analysis of the rule of compatibility of toxic Chinese medicine based on data mining

目的基于数据挖掘探讨毒性中药的使用情况以及高频毒性中药的配伍减毒规律,为规避药物不良反应提供参考。方法以“配伍减毒”为主题,在中国知网、维普、万方等数据库检索界面进行检索,人工筛选文献,用SPSS Clementine 12.0对其进行频数分析,使用Cytoscape软件进行网络可视化展示。结果归纳总结符合纳入标准的282篇文献,共涉及45味有毒中药,研究较多的有附子、雷公藤、马钱子和关木通。其中与附子配伍减毒中药有17味,与雷公藤配伍减毒中药有25味,与马钱子配伍减毒中药有19味,与关木通配伍减毒中药有14味,与四者均有配伍具有减毒作用的中药有16味,其中甘草出现频数最高(139次);从中药功效类别看,配伍减毒中药中补虚药和清热药占比最多。结论利用数据挖掘方法探究毒性中药的应用情况和高频毒性中药配伍减毒用药规律,对毒性中药的临床合理应用有一定的指导意义;对开展配伍减毒机制和用药规律研究提供指导,为国家自然科学“有毒”中药重大科学问题研究提供参考。

Objective To explore the application of toxic traditional Chinese medicine(TCM)and the compatibility of high-frequency toxic TCM based on data mining,therefore to provide reference for avoiding adverse drug reactions.Methods The keywords of"compatibility attenuation"was used to search the information from CNKI,VIP,WANFANG Date and other databases.Also,the related literatures were manually screened.SPSS Clementine 12.0 was used for frequency analysis,and Cytoscape software was used for network visual display.Results A total of 45 toxic Chinese herbal medicines were summarized from 282 literatures according to the inclusion criteria;and the most studied toxic Chinese herbal medicines were Aconiti,Tripterygium wilfordii,Strychni Semen and Aconitum sinensis.For example,there were 17 kinds of attenuated Chinese herbal medicines compatible with Aconiti,25 kinds of attenuated Chinese herbal medicines compatible with Tripterygium wilfordii,19 kinds of attenuated Chinese herbal medicines compatible with Strychnos,14 kinds of attenuated Chinese herbal medicines compatible with aconite,16 kinds of attenuated Chinese herbal medicines compatible with the above 4 herbs;Among them,Glycyrrhizae has the highest frequency(139 times).From the view point of traditional Chinese medicine classification,the tonic medicine and antipyretic medicine accounted for the most.Conclusion The use of data mining methods to explore the application of toxic Chinese medicines and the rules of high-frequency toxic Chinese medicine compatibility and attenuating drugs has certain guiding significance for the rational clinical application of toxic traditional Chinese medicines.This study provides guidance for the research on compatibility and attenuating mechanisms,and provides reference for research on major scientific issues of toxic traditional Chinese medicine.
