摘要
通过对染镧仔鼠海马皮质组织细胞中活性氧簇(ROS)与丙二醛(MDA)含量的检测,探讨镧暴露对程序性坏死相关氧化应激指标的影响。将Wistar雌性大鼠随机分为对照、低、中和高剂量染镧组。孕鼠通过自由饮水的方式进行染镧,仔鼠断乳前通过吸吮母乳染镧,断乳后通过自由饮水染镧。自出生至生后49 d记录染镧仔鼠体质量,之后利用Morris水迷宫实验检测仔鼠学习记忆能力、HE染色观察仔鼠皮质中神经细胞的变化、免疫测定法测定皮质组织中MDA及ROS的含量。结果显示:与对照组相比,染镧仔鼠皮质系数逐渐增高,皮质细胞数量逐渐减少;染镧仔鼠穿越原隐藏平台位置的次数及第四象限持续时间均减少;染镧仔鼠皮质组织中MDA与ROS的含量增高。可见,染镧使仔鼠海马神经细胞氧化应激增强、海马皮质发育受损并影响仔鼠的学习记忆能力。
The effects of lanthanum exposure on oxidative stress related to programmed necrosis were investigated by detecting the contents of reactive oxygen species(ROS)and malondialdehyde(MDA)in hippocampal cortex cells of lanthanum-exposed rats.Female Wistar rats were randomly divided into control,low,medium and high-dose lanthanum-infected groups.The pregnant rats were infected with Lanthanum by drinking water,the offspring rats in sucking period were infected by breast milk and those after weaning by drink water.The body mass of the offspring rats was recorded till the 49th day after birth.The learning and memory ability of the offspring rats was detected by Morris water maze test.The changes of nerve cells and the content of MDA and ROS in the cortex of them were measured by HE staining and immunoassay,respectively.The results showed that compared with the control group,the cortical coefficient of the La-exposed rats gradually increased and the number of cortical cells gradually decreased.The number of times and duration of the fourth quadrant of the La-infected group crossing the original hidden platform decreased;the contents of MDA and ROS in the cortex of La-exposed rats were increased.Therefore,lanthanum exposure increases the oxidative stress in hippocampal neurons of the offspring rats,damages their hippocampal cortex development and harms their learning and memory ability.
