摘要
研究旨在通过线粒体DNA(mitochondrial DNA,mtDNA)D-loop区分子标记研究清远麻鸡的遗传多样性。试验通过PCR扩增和直接测序的方法对80只清远麻鸡的线粒体DNA D-loop区的全序列进行检测,并与红色原鸡进行系统进化分析。研究结果表明:清远麻鸡mtDNA D-loop区序列长度在1231~1232 bp之间,单倍型多样性、核苷酸多样性和平均核苷酸差异分别为0.861、0.00606和7.456。80条序列共发现30个多态位点,定义了18种单倍型,归属可分为A、B、C、D和E 5个单倍型类群。清远麻鸡在mtDNA水平上具有很高的遗传多样性,有多个母系来源。研究结果为清远麻鸡种质资源遗传评估、有效保种和选育利用提供了理论依据。
The aim of this study was to investigate the genetic diversity of Qingyuan partridge chicken by genetic mtDNA D-loop molecular markers.Eighty Qingyuan partridge chickens were tested by PCR amplification and direct sequencing,then the phylogenetic analysis was performed with Red Jungle fowl.The D-loop region of Qingyuan partridge chicken was 1231 to1232 bp in length.The average haplotype diversity,nucleotide diversity and nucleotide differences were 0.861,0.00606,and7.456,respectively.Thirty variable sites that defined eighteen haplotypes were observed.Phylogenetic analysis showed that Qingyuan partridge chicken was clustered into five haplogroups(A,B,C,D and E).It was concluded that there were highly polymorphic and multiple maternal origins at mitochondrial DNA levels for Qingyuan partridge chicken.Therefore,this work will provide the theoretical basis for further evaluation studies,preservation and utilization of Qingyuan partridge chicken.
