摘要
为探究镉(Cd)对鸡软骨细胞的毒性作用,研究在体外培养鸡软骨细胞的基础上添加不同浓度Cd(0、0.2、2、5、10、20μmol/L)处理后观察鸡软骨细胞活力、形态、软骨细胞分化标志基因表达、线粒体膜电位以及凋亡情况。结果显示:5μmol/L以上浓度Cd均能抑制软骨细胞活力,使细胞表面绒毛状突起减少;2μmol/L以上Cd组细胞核出现裂解,线粒体膜电位显著下降,凋亡相关蛋白caspase裂解活化,细胞凋亡率剂量依赖性增加,软骨细胞分化标志基因Acan、Col2a1、Sox9 mRNA表达水平剂量依赖性下调。结果表明,Cd能抑制鸡软骨细胞活力和分化,促进鸡软骨细胞凋亡,为Cd对家禽软骨组织毒性作用的研究奠定基础。
In order to identify the toxic effect of cadmium(Cd)on chicken chondrocytes,chicken chondrocytes were cultured in vitro at different concentrations of Cd(0,0.2,2,5,10,20μmol/L),and the viability,morphology,chondrocyte differentiation marker genes expression,mitochondrial membrane potential and apoptosis of chicken chondrocytes were observed.The results showed that Cd inhibited the activity of chondrocytes,and the villus-like projections on the cell surface decreased when Cd content was more than 5μmol/L.Treatment with more than 2μmol/L Cd were characterized by cell nucleus cleavage,significantly decreased mitochondrial membrane potential,apoptosis-related protein caspase activation,increased cell apoptosis rate,and decreased expression levels of chondrocyte differentiation marker genes Acan,Col2 a1 and Sox9 in a dose-dependent manner.These results confirmed that Cd could inhibit the activity,differentiation of chicken chondrocytes and promote the apoptosis of chicken chondrocytes,which laid a foundation for the study of the toxic effect of Cd on poultry cartilage.
