亚甲基蓝在鲤体内消除规律研究

摘要 为探究常规用药条件下亚甲基蓝及其代谢物在水产动物体内的分布和消除规律,以鲤(Cyprinus carpio)为研究对象,以质量浓度为15 mg/L的亚甲基蓝溶液浸泡试验鱼0.5 h后,将其饲养于水族箱中,采用高效液相色谱-质谱串联法检测亚甲基蓝及其代谢物在鲤各组织中的质量浓度,采用PK Solver药动学药效学数据处理软件对数据进行分析。结果显示,鲤血液中亚甲基蓝、天青B的达峰时间均为0.083 h,峰浓度值C;分别为61.59、4.97μg/L。亚甲基蓝、天青A、天青B在鲤肝胰脏中的质量浓度高于在肌肉组织中的,第30天时仍能在肝胰脏中检测到,说明亚甲基蓝及其代谢物在肝胰脏中易蓄积。研究结果表明,亚甲基蓝可广泛分布在鲤鱼体内,具有较高的生物利用率,在血浆中消除较快,但在组织中消除较慢。 In order to explore the distribution and elimination of methylene blue and its metabolites in aquatic animals under the condition of conventional medication, high performance liquid chromatography-mass spectrometry tandem method was used to detect the changes of methylene blue concentration in plasma, hepatopancreas and muscle of carp.In this study, Cyprinus carpio was immersed in 15 mg/L methylene blue solution for 0.5 h, and then cultured in an aquarium.PK solver pharmacokinetic pharmacodynamics data processing software was used to analyze the data.The results showed that the peak time(T;) of methylene blue and azure B in the blood of carp were 0.083 h, and the peak concentration(C;) was 61.59 and 4.97 μg/L,respectively.The concentrations of methyl blue, azure A and azure B in hepatopancreas of carp were higher than those in muscle tissue, and they could still be detected in hepatopancreas on day 30,indicating that methylene blue and its metabolites were easy to accumulate in the hepatopancreas.These results indicated that methylene blue could be widely distributed in carp with high bioavailability, and was eliminated quickly in plasma but relatively slower in tissues.
