莲心碱对大鼠非酒精性脂肪肝组织的保护作用及机制研究

Protective Effect and Mechanism of Liensinine on Nonalcocholic Fatty Liver in Rats

目的:研究莲心碱对非酒精性脂肪肝(NAFLD)肝细胞炎症以及NF-κB、TNF-α凋亡相关蛋白表达的影响,探讨莲心碱对NAFLD的治疗作用和机制。方法:高脂饲料喂养大鼠8周构建NAFLD模型,模型构建成功后随机分为正常空白组、高脂模型组、莲心碱低剂量组和高剂量组,每组8只。喂养至12周末麻醉处死后取材,HE染色观察大鼠肝组织病理学改变情况,检测肝组织血清谷丙转氨酶(ALT)、总胆固醇(TC)、甘油三酯(TG)表达水平,NF-κB、TNF-α表达情况。结果:高脂模型组NF-κB、TNF-α、ALT、TC、TG水平显著高于正常对照组(P<0.05)。相比高脂模型组,莲心碱高、低剂量组水平降低(P<0.05),且剂量越高降低越明显。结论:莲心碱可减少NF-κB、TNF-α的产生,发挥抗炎作用,从而对NAFLD引起的血脂增高和肝功能损伤起到一定保护和治疗作用。

Objective To observe the effects of Liensinine on the expression of NF-κB and TNF-αproteins related to inflammation and apoptosis in hepatocytes of rats with non-alcoholic fatty liver disease(NAFLD),and to explore the protective effect and mechanism of liensinine in the pathogenesis of NAFLD.MethodsNAFLD model was established in rats fed with high-fat diet for 8 weeks.After the model was established,rats were randomly divided into control group,high-fat model group,low-dose group and high-dose group with 8 rats in each group.The rats were sacrificed at the end of 12 weeks after feeding.The pathological changes of liver were observed by HE staining.The expression levels of alanine aminotransferase(ALT),total cholesterol(TC),triglyceride(TG),and the expressions of NF-κB and TNF-αin serum of liver tissues were detected.ResultsCompared with the control group,levels of NF-κB,TNF-α,ALT,TC and TG in high fat model group were significantly increased(P<0.05).Compared with the high-fat model group,the levels of lieninine in high-dose and low-dose groups were decreased(P<0.05).The higher the dose,the more significant the decrease was.Conclusions Lieninine can reduce the production of NF-κB and TNF-α,and play an anti-inflammatory effect,thus playing a protective and therapeutic role in the increase in blood lipid and liver function injury caused by NAFLD.
