摘要
目的研究miR-622抑制物(miR-622 inhibitor)对人乳腺癌细胞MCF-7增值、迁移与侵袭的影响。方法待对数生长期状态良好的人乳腺癌细胞MCF-7生长融合度达30%~50%时,使用Ribofactamine转染试剂将miR-622 inhibitor及miR-622阴性对照(miR-NC)分别转染到MCF-7细胞中,分组为miR-622 inhibitor组、miR-NC组及control组。采用qRT-PCR法分别检测3组MCF-7细胞中miR-622的表达水平,Western blot检测3组细胞中EYA1蛋白表达水平。同时采用CCK-8实验、伤口愈合实验、Transwell小室实验分析miR-622 inhibitor对MCF-7细胞增殖、迁移和侵袭的影响。结果miR-622 inhibitor组中miR-622相对表达量低于control组及miR-NC组,EYA1蛋白相对表达水平高于control组和miR-NC组,差异有统计学意义(P<0.05);实验结果显示,miR-622 inhibitor组增殖能力、迁移率及侵袭细胞数均低于control组及miR-NC组,差异有统计学意义(P<0.05)。结论miR-622低表达可能通过上调EYA1通路,实现降低乳腺癌细胞的增殖、迁移和侵袭水平,抑制了癌细胞的转移。
Objective To study the effect of miR-622 inhibitor on the proliferation,migration and invasion of human breast cancer MCF-7 cells.Methods miR-622 inhibitor and miR-622 negative control(miR-NC)were transfected into MCF-7 cells by Ribofactamine transfection reagent when the growth fusion degree of human breast cancer cell MCF-7 in good logarithmic growth phase was 30%-50%.The cells were divided into miR-622 inhibitor group,miR-NC group and control group.qRT-PCR was used to detect the expression level of miR-622 in MCF-7 cells of the three groups,and Western blot was used to detect the expression level of EYA1 protein in the three groups.At the same time,CCK-8 experiment,wound healing experiment and Transwell chamber experiment were used to analyze the effects of miR-622 inhibitor on the proliferation,migration and invasion of MCF-7 cells.Results The relative expression level of miR-622 in the miR-622 inhibitor group was lower than that in the control group and miR-NC group,and the relative expression level of EYA1 protein was higher than that in the control group and miR-NC group,and the difference was statistically significant(P<0.05).CCK-8 results showed that after 12 h,the proliferation ability,migration rate and the number of invasive cells in the miR-622 inhibitor group were lower than those in the control group and the miR-NC group,and the difference was statistically significant(P<0.05).Conclusion The low expression of miR-622 may reduce the proliferation,migration and invasion of breast cancer cells and inhibit the metastasis of cancer cells by up-regulating the EYA1 pathway.
