miR-103a-3p靶向PTEN基因促进血管瘤内皮细胞增殖、迁移及侵袭 被引量:3

摘要 目的:探讨miR-103a-3p对人血管瘤内皮细胞(HemEC)增殖、迁移和侵袭的影响及作用机制。方法:分别以正常皮肤组织和人脐静脉内皮细胞(HUVEC)为对照,RT-qPCR检测人血管瘤组织和HemEC中miR-103a-3p和PTEN mRNA表达,Western blot检测PTEN蛋白表达。HemEC分为anti-miR-130b-3p组、anti-miR-NC组、pcDNA组、pcDNA-PTEN组、anti-miR-130b-3p+si-con组、anti-miR-130b-3p+si-PTEN组,MTT法检测各组细胞增殖,Transwell检测各组细胞迁移和侵袭。双荧光素酶报告基因检测实验验证miR-103a-3p与PTEN的靶向关系。结果:与正常皮肤组织比较,血管瘤组织中miR-103a-3p表达水平显著升高(P<0.05),PTEN的mRNA和蛋白表达水平显著降低(P<0.05)。与HUVEC比较,HemEC中miR-103a-3p表达显著升高(P<0.05),PTEN的mRNA和蛋白表达均显著降低(P<0.05)。与anti-miR-NC组比较,anti-miR-103a-3p组HemEC的OD值、迁移和侵袭细胞数均显著降低(P<0.05)。与pcDNA组比较,pcDNA-PTEN组HemEC的OD值、迁移和侵袭细胞数均显著降低(P<0.05)。miR-103a-3p在HemEC中负调控PTEN表达。与anti-miR-103a-3p+si-NC组比较,anti-miR-103a-3p+si-PTEN组HemEC的OD值显著升高、迁移和侵袭细胞数均显著升高(P<0.05)。结论:miR-103a-3p在血管瘤组织和HemEC中呈高表达,抑制其表达通过负调控PTEN降低HemEC的增殖、迁移和侵袭能力。 Objective:To investigate the effects and mechanism of miR-103a-3p on the proliferation,migration and invasion abilities of hemangioma endothelial cells(HemEC).Methods:Using normal skin tissue and human umbilical vein endothelial cells(HUVEC)as controls,RT-qPCR was used to detect expressions of miR-103a-3p and PTEN mRNA in human hemangioma tissue and HemEC,and Western blot was used to detect the expression of PTEN protein. HemEC were divided into anti-miR-130 b-3 p group,anti-miR-NC group,pcDNA group,pcDNA-PTEN group,anti-miR-130 b-3 p+si-con group and anti-miR-130 b-3 p+si-PTEN group,then MTT method was used to detect cell proliferation in each group,and Transwell was used to detect cell migration and invasion in each group. The dual luciferase reporter gene detection experiment verified the targeting relationship between miR-103a-3p and PTEN.Results:Compared with normal skin tissues,the expression level of miR-103a-3p in hemangioma tissues was significantly increased(P<0.05),and the expression levels of PTEN mRNA and protein were significantly reduced(P<0.05). Compared with the anti-miR-NC group,the OD value of HemEC and the number of cells that migrated and invaded in the anti-miR-103a-3p group were significantly decreased(P<0.05). Compared with the pcDNA group,the OD value of HemEC and the number of cells that migrated and invaded in the pcDNA-PTEN group were significantly decreased(P<0.05). miR-103a-3p negatively regulated PTEN expression in HemEC. Compared with the anti-miR-103a-3p+si-NC group,the OD value of HemEC and the number of cells that migrated and invaded in the anti-miR-103a-3p+si-PTEN group were significantly elevated(P<0.05).Conclusion:miR-103a-3p is highly expressed in hemangioma tissue and HemEC,and inhibiting its expression reduces the proliferation,migration and invasion of HemEC by negatively regulating PTEN.
