摘要 目的:检测脓毒症中CCL15的含量,探究CCL15在脓毒症免疫病理中的作用和机制。方法:选取健康对照者和脓毒症患者各30例,通过盲肠结扎穿刺建立动物模型(CLP),ELISA检测患者和小鼠血清中CCL15含量;给脓毒症小鼠补充重组人CCL15蛋白或PBS,探究CCL15对生存率的影响;通过HE染色和生化指标检测评估器官损伤;通过流式细胞术检测吞噬细胞变化,探究CCL15对细胞吞噬杀伤的影响。结果:脓毒症患者和脓毒症小鼠的CCL15水平显著升高;在实验性脓毒症中,补充CCL15蛋白可以显著提高生存率,改善器官损伤,促进细菌清除。CCL15处理可以促进腹腔灌洗液(PLF)中巨噬细胞募集。结论:CCL15促进细菌清除,改善器官损伤,在脓毒症中发挥免疫保护作用,并改善生存率。 Objective:To investigate the CCL15 expression in sepsis and explore the mechanism underlying the effect.Methods:The study enrolled 30 healthy individuals and 30 human sepsis patients.Animal models were established by cecal ligation and puncture(CLP),and ELISA was used to detect serum CCL15 levels of human and mouse.Recombinant human CCL15 protein or PBS was used to explore the effect of survival rate.HE staining and some biochemical indexes were used to evaluate pathologic injury.The changes of phagocytes were detected by flow cytometry,and the effects of CCL15 on cell phagocytosis and killing were investigated.Results:CCL15 levels were significantly elevated in human and murine sepsis.CCL15 administration significantly improved survival,bacterial clearance,and organ damage.An increased macrophage influx was developed of CCL15-treated mice in PLF.Conclusion:CCL15 promotes bacterial clearance and improves organ damage.It plays an immune protective role in sepsis,and improves survival.
