摘要
前列腺癌(PCa)是男性泌尿生殖系统常见的恶性肿瘤之一,其治疗方法以外科手术、放化疗及内分泌治疗为主,预后较差。m6A是真核生物中最常见的RNA修饰形式,其过程由甲基转移酶、去甲基化酶和甲基化结合蛋白参与完成,是一种可逆的动态修饰。m6A修饰在参与RNA剪切、转运、翻译和稳定性等方面发挥着重要作用,其相关蛋白包括METTL3、FTO、YTHDF2等与PCa的发生发展密切相关,针对m6A甲基化修饰及其相关蛋白为靶点的治疗方法,可能成为治疗前列腺癌的一个潜在的新方向。本文就m6A与前列腺癌的作用机制和最新的研究进展作一综述。
Prostate cancer(PCa)is one of the most common malignancies of male genitourinary system.The treatment methods are mainly surgery,chemoradiotherapy and endocrine therapy,but the prognosis is poor.N6-methyladenosine(m6A)is the most common form of RNA modification in eukaryotes,and its process is completed by methyltransferase,demethylase and methylated binding protein,which is a reversible dynamic modification.m6A modification plays an important role in RNA splicing,transport,translation and stability.Its related proteins include METTL3,FTO,YTHDF2,and so on,which are closely related to the development and progression of prostate cancer.Treatments targeting m6A methylation modification and its related proteins may be a potential new direction for the treatment of PCa.This paper focuses on the mechanism of m6A,prostate cancer and the latest research progress.
