冬凌草甲素纳米脂质体对嗜水气单胞菌生物膜的抑制活性研究 被引量:3

摘要 为探究冬凌草甲素纳米脂质体对嗜水气单胞菌生物膜形成的影响,该研究首先采用二倍稀释法测定冬凌草甲素纳米脂质体对嗜水气单胞菌的最小抑菌浓度(minimum inhibitory concentration,MIC),之后采用结晶紫法、四甲基偶氮唑盐微量酶反应比色法[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction,MTT]、荧光显微观察及电镜扫描等方法评价冬凌草甲素纳米脂质体在不同质量浓度时对嗜水气单胞菌生物被膜形成的抑制作用及清除效果。试验结果表明,冬凌草甲素纳米脂质体对嗜水气单胞菌的MIC为62.5μg/mL;冬凌草甲素纳米脂质体在亚抑菌浓度时对嗜水气单胞菌生物膜形成的抑制作用和清除效果不明显;当浓度大于MIC时,其对嗜水气单胞菌生物膜的形成具有显著的抑制作用和清除效果,并且可以显著抑制该菌的代谢活性。 To explore the effect of oridonin nanoliposomes on Aeromonas hydrophila biofilm formation,the minimum inhibitory concentration(MIC)of oridonin nanoliposomes on A.hydrophila was determined using a double dilution method.Dose dependent inhibition and scavenging effects of oridonin nanoliposomes on A.hydrophila biofilm formation was evaluated by crystal violet staining,3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction(MTT)assay,fluorescence microscopy,and scanning electron microscopy.The MIC of oridonin nanoliposomes against A.hydrophila was 62.5μg/mL.Inhibition and scavenging effects of oridonin nanoliposomes on A.hydrophila biofilm formation were not obvious at sub-inhibitory concentrations.At concentrations greater than MIC,they displayed significant inhibition and scavenging effects on A.hydrophila biofilm formation,and significantly inhibited the metabolic activity of the strain.
