黄芪甲苷葡萄糖注射液抗缺血性心肌损伤的实验研究 被引量:2

摘要 目的观察黄芪甲苷葡萄糖注射液对心肌缺氧、缺血动物模型抗心肌损伤的作用及其机制。方法比较常压耐缺氧实验中溶媒组、维拉帕米组(2 mg/kg)及黄芪甲苷葡萄糖注射液高剂量组(以下简称高剂量组,8 mg/kg)、黄芪甲苷葡萄糖注射液中剂量组(以下简称中剂量组,4 mg/kg)、黄芪甲苷葡萄糖注射液低剂量组(以下简称低剂量组,2 mg/kg)小鼠停止呼吸的时间;比较异丙肾上腺素(ISO)诱导的心肌缺血小鼠耐缺氧实验中正常组、溶媒组、维拉帕米组(2 mg/kg)、高剂量组(8 mg/kg)、中剂量组(4 mg/kg)、低剂量组(2 mg/kg)小鼠停止呼吸的时间;比较夹闭气管致大鼠心电消失实验溶媒组、维拉帕米组(1 mg/kg)、高剂量组(4 mg/kg)、中剂量组(2 mg/kg)、低剂量组(1 mg/kg)大鼠心电消失时间。通过ISO诱导建立大鼠心肌缺血模型,观察并记录溶媒组、硝酸异山梨酯组(2 mg/kg)、高剂量组(4 mg/kg)、中剂量组(2 mg/kg)、低剂量组(1 mg/kg)大鼠心电图总ST段偏移幅度(∑ST)。通过结扎冠状动脉左前降支建立心肌缺血大鼠模型,比较假手术组、溶媒组、硝酸异山梨酯组(2 mg/kg)、高剂量组(4 mg/kg)、中剂量组(2 mg/kg)、低剂量组(1 mg/kg)大鼠全血黏度(低切、中切、高切)、血浆黏度、红细胞比容。结果与溶媒组相比,常压缺氧实验和心肌缺血小鼠耐缺氧实验中维拉帕米组、高剂量组和中剂量组小鼠呼吸时间均显著延长(P<0.05),气管夹闭心电消失实验中维拉帕米组、高剂量组和中剂量组大鼠心电消失时间均显著延长(P<0.05),心电ST段总偏移实验中硝酸异山梨酯组、高剂量组和中剂量组大鼠∑ST均明显降低(P<0.05),血液流变学实验中硝酸异山梨酯组、高剂量组和中剂量组大鼠全血黏度(低切、中切、高切)、血浆黏度和红细胞比容均降低(P<0.05)。结论黄芪甲苷葡萄糖注射液能够改善动物模型的心肌损伤状态,抗心肌损伤作用可能与增强心肌细胞耐缺氧能力、降低血液流变学参数有关。 Objective To observe the effects and mechanism of astragaloside glucose injection for myocardial hypoxia and anti-myocardial injury in ischemic model.Methods The breathless time of rats in the vehicle group,verapamil group(2 mg/kg),high-dose astragaloside glucose injection group(hereinafter referred to as the high-dose group,8 mg/kg),medium-dose astragaloside glucose injection group(hereinafter referred to as the medium-dose group,4 mg/kg),low-dose astragaloside glucose injection group(hereinafter referred to as the low-dose group,2 mg/kg)was compared in hypoxia tolerance experiments under normal pressure;the breathless time of mice in the normal group,vehicle group,verapamil group(2 mg/kg),high-dose group,(8 mg/kg),medium-dose group(4 mg/kg),low-dose group(2 mg/kg)was compared through isoprenaline(SO)-induced hypoxia tolerance experiments for myocardial ischemia mice.The disappearance time of electrocardiographic(ECG)beats of rats in the vehicle group,verapamil group(1 mg/kg),high-dose group(4 mg/kg),medium-dose group(2 mg/kg),low-dose group(1 mg/kg)was compared through occlusion of trachea-induced ECG disappearance experiences for rats.Myocardial ischemia model was established by ISO induction,and total ECG ST offset(ΣST)of rats in the vehicle group,isosorbide dinitrate group(2 mg/kg),high-dose group(4 mg/kg),medium-dose group(2 mg/kg),low-dose group(1 mg/kg)were observed and recorded.The myocardial ischemia rat model was established by coronary left descending artery ligation,and the whole blood viscosity(WBV)(low,medium and high shears),plasma viscosity,hematocrit of rats in the sham operation group,vehicle group,isosorbide dinitrate group(2 mg/kg),high-dose group(4 mg/kg),medium-dose group(2 mg/kg),low-dose group(1 mg/kg)were compared.Results Compared to mice in the vehicle group,the breathless time of mice in the verapamil group,medium-dose group,and high-dose group through the hypoxia tolerance experiments was significantly prolonged(P<0.05);ECG disappearance time of rats in the verapamil group,medium-dose group,and high-dose group through occlusion of trachea-related ECG disappearance experiments for rats was significantly prolonged(P<0.05);ΣST of rats in the isosorbide dinitrate group,medium-dose,and high-dose group through ST total offset experiment of ECG was significantly decreased(P<0.05);WBV(low,medium and high shears),plasma viscosity and hematocrit of rats in the isosorbide dinitrate group,medium-dose,and high-dose group through hemorheology experiments was significantly decreased(P<0.05).Conclusion Astragaloside glucose injection is capable of improving myocardial injury in animal models.The anti-myocardial injury effect may be correlated with enhanced hypoxia tolerance of cardiomyocytes and decreased hemorheological parameters.
