摘要
目的探讨四君子汤(SiJunZi Decoction,SJZD)治疗大鼠阿尔茨海默病的作用机制。方法将60只雄性SPF级大鼠随机分为正常组,模型组,双氢麦角碱(dihydroergoline,DHE)组,SJZD低、中、高治疗组。采用Morris水迷宫实验检测大鼠记忆功能的变化;ELISA法检测IL-1β、IL-6及IL-10含量;免疫组化法检测caspase11蛋白表达变化;Western blot法检测海马组织NLRP3、caspase-1、caspase-4、caspase-5蛋白表达。结果与正常组比较,模型组大鼠潜伏逃避期时间增加(P<0.01)、穿越次数减少(P<0.01),IL-1β、IL-6含量升高(P<0.01),IL-10含量降低(P<0.01),NLRP3、caspase-1、caspase-4、caspase-5、caspase-11(P<0.01)表达上调;DHE与模型组比较,潜伏逃避期减少(P<0.01),穿越次数增加(P<0.01),IL-1β减少(P<0.01),NLRP3、caspase-1表达下调(P<0.01)。SJZD中剂量组和高剂量组与模型组比较,潜伏逃避期时间减少(P<0.01)、穿越次数增加(P<0.01),caspase-11蛋白表达下调;SJZD低剂量组、中剂量组和高剂量组与模型组比较IL-1β、IL-6含量降低(P<0.01),IL-10含量升高(P<0.01),NLRP3、caspase-1、caspase-4、caspase-5蛋白表达下调(P<0.01)。结论四君子汤治疗阿尔茨海默病可能与纠正中枢海马相关炎症因子紊乱及调控NLRP3有关。
To evaluate the therapeutic effect of Sijunzi Decoction(SJZD)on Alzheimer’s disease by regulating central inflammatory factors in rats,total of 60 male SPF rats were recruited and randomly divided into normal group,model group,dihydroergoline(DHE)group and SJZD low,medium and high doses groups,with 10 rats in each group.The changes in memory function of rats were detected by Morris water maze test,while the levels of IL-1β,IL-6 and IL-10 were detected by ELISA;the expression changes of caspase 11 protein was detected by immunohistochemistry;the protein expressions of NLRP3,caspase-1,caspase-4 and caspase-5 in hippocampus were detected by Western blotting.Data showed that compared with the normal group,the latency escape time of the model group increased(P<0.01),the number of crossing decreased(P<0.01),and the levels of IL-1β and IL-6 increased(P<0.01),the content of IL-10 decreased(P<0.01),the expression of NLRP3,caspase-1,caspase-4,Caspase-5 and caspase-11 increased(P<0.01);compared with the model group,the latent escape time of DHE group decreased(P<0.01),the number of crossing increased(P<0.01),the expression of IL-1β(P<0.01),and the expression of NLRP3 and caspase-1 decreased(P<0.01).Compared with the model group,the latency escape time decreased(P<0.01),the number of crossing increased(P<0.01),and the expression of caspase-11 protein decreased in the middle dose group and high dose group of SJZD.Compared with the model group,theexpression of IL-1βin all groups of SJZD weredecreased,the content of IL-6 was decreased(P<0.01),the content of IL-10 was increased(P<0.01),and theexpression of NLRP3,caspase-1,caspase-4 and Caspase-5 were decreased(P<0.01).In conclusion,Sijunzi Decoction in the treatment of Alzheimer’s disease maybe related to correcting the disorder of central hippocampal inflammatory factors and regulating NLRP3.
