益黄散治疗小儿慢性腹泻的网络药理学研究 被引量:1

摘要 目的 采用网络药理学探讨益黄散治疗小儿慢性腹泻的分子作用机制,为临床用药提供依据。方法 通过中药系统药理学数据库与分析平台(TCMSP)挖掘益黄散药物有效成分,导入PubChem、SwissTargetPrediction数据库预测有效成分潜在靶点;在GeneCards数据库中检索小儿慢性腹泻靶点,将有效成分靶点与疾病靶点取交集,并绘制中药-有效成分-疾病网络。通过STRING11.0数据库构建蛋白相互作用网络,运用Cytoscape 3.8.1软件Network analyzer功能整合并构建药物成分-靶点-疾病的多层次网络关系。通过DAVID数据库对交集靶点进行GO功能富集及KEGG通路富集分析。结果 收集并筛选益黄散有效成分19个、药物靶点541个,小儿慢性腹泻疾病靶点2574个,二者交集靶点134个,其中PIK3CA、STAT3、VEGFR、AKT1、EGFR等基因靶点相互作用明显。GO功能富集得到生物过程条目358个,细胞成分条目4个,分子功能条目42个;KEGG通路富集共计82条信号通路,主要调控癌症途径、癌症中蛋白聚糖、Ras信号通路、PI3K-Akt信号通路、局灶性粘连、MAPK信号通路等。结论 本研究揭示了益黄散在小儿慢性腹泻治疗中多途径、多靶点、多成分的作用特点,可为后续研究提供理论基础。 Objective To study the molecular mechanism of Yihuang Powder for the treatment of infantile chronic diarrhea with network pharmacology;To provide basis for clinical medication. Methods The active components of Yihuang Powder were dug from TCMSP, and then PubChem and SwissTargetPrediction databases were imported to predict potential targets of active components;the targets of infantile chronic diarrhea were searched in GeneCards database, the intersection of active component targets and disease targets was taken, and the Chinese materia medica-active components-disease network was drawn. The protein-protein interaction network was constructed by STRING 11.0 database and Cytoscape 3.8.1 Network analyzer was used to integrate and construct the multi-level network relationship of medicine composition-target-disease. GO function enrichment and KEGG pathway enrichment analysis of intersection targets were carried out through DAVID database. Results Totally 19 active components, 541 medicine targets of Yihuang Powder, 2 574 disease targets of infantile chronic diarrhoea, and 134 intersection targets of the two were collected and screened, among which the gene targets of the PIK3 CA, STAT3, VEGFR, AKT1, and EGFR were obvious. GO function enrichment resulted in 358 biological process entries, 4 cell component entries and 42 molecular function entries;KEGG pathway was enriched with 82 signaling pathways, mainly regulating cancer pathway, proteoglycan in cancer, Ras signaling pathway, PI3 K-Akt signaling pathway, focal adhesion, MAPK signaling pathway, etc.Conclusion This study reveals the multi-channel, multi-target and multi-component characteristics of Yihuang Powder in the treatment of infantile chronic diarrhea, so as to provide a theoretical basis for follow-up research.
