摘要
植物乳杆菌在维持人体健康方面起着重要的作用,但在生产加工过程中以及在胃肠道内消化后,活菌数降低,严重影响益生效果。本研究采用微滴喷射系统装置制备植物乳杆菌微胶囊,以微胶囊包埋率为评价指标,确定海藻酸盐、低酯果胶和卵磷脂复合壁材的最佳浓度组合。结果表明,海藻酸钠质量浓度1%kg/L,卵磷脂质量浓度1%kg/L,低酯果胶质量浓度0.75%kg/L,氯化钙浓度0.4 mol/L时,微胶囊的包埋率达到最高89.05%。微胶囊经模拟胃液处理2 h后菌体存活率为62.3%,活菌数为4.38×10^(9)cfu/mL;经模拟肠液处理2 h后几乎完全释放,活菌数达7.14×10^(9)cfu/mL。以海藻酸盐、低酯果胶和卵磷脂为复合壁材,微滴喷射法制备植物乳杆菌微胶囊减少了菌体受环境的侵害,提高了菌体在宿主体内的存活率。
Lactobacillus plantarum plays an important role in maintaining human health.However,during the production process and after digestion in the gastrointestinal tract,the reduction in the number of viable bacteria seriously affects the probiotic effect.In this study,the micro-droplet spray system was used to prepare Lactobacillus plantarum microcapsules.Alginate,low-ester pectin and lecithin were used as composite wall materials,and the optimal concentration combination of wall materials was determined based on the microcapsule embedding rate.The results show that when 1%kg/L sodium alginate,1%kg/L lecithin,0.75%kg/L low-ester pectin,and 0.4 mol/L calcium chloride are used as wall materials,the highest microcapsule embedding rate is 89.05%.After the microcapsules are treated with simulated gastric juice for 2 h,the survival rate of the bacteria is 62.3%,and the number of viable bacteria is 4.38×10^(9)cfu/mL.After being treated with simulated intestinal juice,the microcapsules almost completely release after 2 h.The number of viable bacteria reaches 7.14×10^(9)cfu/mL.Microencapsules of Lactobacillus plantarum by micro-droplet spray reduces the damage of the bacteria by the environment and improves the survival rate of the bacteria in the host.
