Expression and significance of Smad4 in placental tissue of early onset preeclampsia

摘要 目的探讨早发型子痫前期胎盘组织中信号转导因子(Smad4)的表达及其可能的作用。方法采用实时荧光定量PCR(q RT-PCR)、蛋白印迹法(Western blot)、免疫组化检测30例早发型子痫前期和30例健康孕妇胎盘组织中Smad4的表达情况;Transwell和划痕实验检测人绒毛膜滋养层细胞系(HTR-8/SVneo-细胞)的侵袭和迁移情况。结果在早发型子痫前期胎盘组织中Smad4 mRNA和蛋白水平均显著增高,差异均有统计学意义(P<0.01);与阴性对照组(si-Smad4 NC组)相比,沉默Smad4后,HTR-8/SVneo细胞侵袭力和迁移率显著增高,差异有统计学意义(P<0.01)。结论在早发型子痫前期胎盘组织中Smad4表达明显升高,并通过影响滋养细胞生物学功能参与子痫前期的发生发展。 Objective To investigate the expression of Smad4 in placental tissue of early onset preeclampsia and its possible role.Method Smad4 expression was detected by RT-PCR,Western blot and immunohistochemistry in placental tissues of 30 cases of early onset preeclampsia and 30 healthy parturients.Transwell and scratch assay were used to detect the invasion and migration of trophoblast cell line HTR-8/SVneo cells.Result Smad4 mRNA and protein levels in placental tissues of early onset preeclampsia were significantly increased(P<0.01);Compared with the siSmad4 NC group,the invasion and mobility of HTR-8/SVneo cells were significantly increased after Smad4 silencing,and the difference was statistically significant(P<0.01).Conclusion Smad4 expression is significantly increased in placental tissue of early onset preeclampsia,which may be involved in the occurrence and development of preeclampsia by affecting the biological function of trophoblast cells.
