摘要
芭蕉芋是一种富含淀粉的具有较高经济价值的多用途作物,而美人蕉属于常见的草本园艺观赏植物,虽然两个物种用途广泛,但是它们的分子机制研究一直没有重要突破。为了加速两者分子育种的发展,本研究利用高通量测序技术对芭蕉芋和美人蕉进行Survey分析,通过生物信息学分析,完成了芭蕉芋和美人蕉全基因组特征评估,获得了基因组大小、杂合度以及GC含量等信息,基于两个基因组的特征比较分析结果完成了芭蕉芋基因组的初步组装。结果表明,通过对芭蕉芋和美人蕉进行高通量测序,分别得到139.30和68.10 Gb高质量的测序数据,总测序深度分别约为140 x和68 x,估算的基因组大小分别约为844.90和836.88 Mb;根据K-mer分析估算全基因组GC含量分别为39.14%和38.48%,杂合度分别为0.63%和1.50%。由此可知,芭蕉芋和美人蕉基因组均属于较为复杂的基因组。对芭蕉芋尝试初步组装后得到总长为555.60 Mb的基因组。本研究获得了芭蕉芋和美人蕉基因组大小和特征等信息,为芭蕉芋和美人蕉的分子育种研究提供了有价值的数据与思路,初步比较了芭蕉芋和美人蕉基因组特征的异同,为后续构建两者全基因组精细图谱以及比较图谱提供了理论基础。
Canna edulis Ker is a kind of multi-purpose crops with high starch content and economic value. Canna indica L. belong to common herb garden ornamental plants. Although the two species have wide applications,there has been no important breakthrough in the molecular mechanism research of these two species. In order to accelerate the development of molecular breeding for them, in this study, high-throughput sequencing technology was used to conduct survey analysis on the two species of Canna edulis and Canna indica. Through bioinformatics analysis, the whole genome characteristics of Canna edulis and Canna indica were evaluated, and the genome size,heterozygosity and GC content were obtained. The genomes were preliminary assembled and comparative analysis was conducted. Based on the results of feature comparison and analysis of the two genomes, the preliminary assembly of the Canna edulis genome was completed. Through high-throughput sequencing, 139.30 and 68.10 Gb high-quality data were obtained in Canna edulis Ker and Canna indica L., with a total sequencing depth of about140 x and 68 x, and the genome sizes of them were estimated to be 844.90 and 836.88 Mb, respectively. Besides,based on K-mer analysis, the GC content of the whole genome were estimated to be 39.14%(Canna edulis Ker)and 38.48%(Canna indica L.), and the heterozygosity were 0.63%(Canna edulis Ker) and 1.50%(Canna indica L.),respectively. Therefore, both Canna edulis Ker and Canna indica L. are complex genomes. A draft genome of Canna edulis Ker was obtained by preliminary assembly with a total length of 555.60 Mb. In this study, the size and characteristics of genomes in Canna edulis Ker and Canna indica L. were obtained, and the genomes were compared, which will provide a basis for the whole genomes fine mapping, and also provide valuable data for molecular breeding for both of them in the future.
