摘要
为建立鱼腥草快繁体系,本研究以鱼腥草种子为实验材料,探究在植物组织培养过程中外植体灭菌、种子萌发、壮苗以及幼苗移栽相关条件。实验结果表明,鱼腥草种子在75%乙醇30 s+0.2%Hg Cl_(2)5 min灭菌后能够在无菌时获得55.26%的萌发率;最优启动培养基为1/2MS+0.5 mg/L GA_(3)+0.2 g/L活性炭粉;幼苗最佳壮苗培养基为1/2MS+0.1 mg/L KT+0.3 mg/L NAA+0.2 mg/L GA_(3)+0.2 g/L活性炭粉;幼苗在移栽前转移至自然光温条件下生长1周,再开盖适应5 d后栽种在浇灌0.2 g/L甲基托布津溶液的苗圃土中可获得最高的成活率86.67%。本研究成功建立了稳定的鱼腥草快繁体系,将为农业生产、生物实验等领域提供技术和材料支持。
In order to establish a rapid propagation system of Houttuynia cordata Thunb.,the relevant conditions of explants sterilization,seeds germination,strong seedlings and seedling transplanting in the process of plant tissue culture were investigated using seeds as the experimental materials.The results showed that the seeds can obtain 55.26%germination rate when sterilized in 75% ethanol for 30 s followed 0.2%Hg Cl_(2)for 5 minutes;the optimal germination medium for seeds is 1/2MS+0.5 mg/L GA_(3)+0.2 g/L activated carbon;the suitable rooting medium is 1/2MS+0.1 mg/L KT+0.3 mg/L NAA+0.2 mg/L GA_(3)+0.2 g/L activated carbon;seedlings were transferred to natural light and temperature to grow for 1 week,and then opened the cover to adapt for 5 days before transplanting.After planted,the highest survival rate was 86.67% in nursery soil watered with 0.2 g/L thiophanate-methyl solution.This study has successfully established a stable rapid propagation system of H.cordata this will provide technical and material support for agricultural production and biological experiments.
