摘要
目的:观察三化汤对缺血性中风模型大鼠血清脂肪酸谱的影响。方法:采用线栓法建立大鼠大脑中动脉阻塞模型(MCAO模型),随机分为模型组、干预组,每组10只。另设正常组、假手术组,每组各10只。正常组、假手术组、模型组灌胃纯净水,干预组灌胃三化汤水煎液,连续给药10 d。10 d后处死大鼠,取血清样本,采用气相色谱-质谱联用技术(GC-MS),以水杨酸甲酯为内标进样测定,按标准曲线法计算血清中脂肪酸的含量。结果:与正常组比较,模型组大鼠血清中C6:0、C8:0、C12:0、C13:0、C16:0、C18:0、C20:0、C10:0、C14:0等饱和脂肪酸(saturated fatty acid,SFA)含量均显著升高(P<0.05或P<0.01);C14:1T、C14:1、C15:1T、C15:1、C16:1T、C16:1、C17:1T、C17:1、C18:1N12T、C18:1N7T、C19:1N12T、C20:1T、C22:1N9T、C22:1N9、C24:1等单不饱和脂肪酸(monounsaturated fatty acid,MUFA)含量均显著升高(P<0.05或P<0.01);反式不饱和脂肪酸(trans unsaturated fatty acids,TRNFA)含量显著升高(P<0.01)。与模型组比较,干预组大鼠血清中C16:0、C18:0、C20:0、C10:0、C14:0等SFA含量均显著降低(P<0.05或P<0.01);C14:1T、C14:1、C15:1、C16:1T、C16:1、C17:1、C18:1N12T、C18:1N9T、C18:1N7T、C20:1T等MUFA含量均显著降低(P<0.05或P<0.01);TRNFA含量显著降低(P<0.05)。结论:缺血性中风模型大鼠血清存在脂肪酸代谢紊乱,尤其是饱和脂肪酸、单不饱和脂肪酸和反式不饱和脂肪酸含量升高明显;三化汤干预后显著改善了其血清中饱和脂肪酸、单不饱和脂肪酸和反式不饱和脂肪酸含量的异常变化,调节紊乱的脂肪酸谱趋于正常。
OBJECTIVE To observe the effects of Sanhua Decoction on serum fatty acid profile of rats with ischemic stroke.METHODS The rat’s middle cerebral artery occlusion model(MCAO model)was established by the thread plug method.The rats were randomly divided into model group and intervention group with10 rats in each group.In addition,a normal group and a sham operation group were set up with10 rats in each group.The normal group,sham operation group and model group were intragastrically administered with purified water,and the intervention group was intragastrically administered with Sanhua decoction for 10 consecutive days.After 10 days,the rats were sacrificed and serum samples were taken.The samples were determined by gas chromatography-mass spectrometry(GC/MS),and methyl salicylate was used as the internal standard.The serum fatty acid content was calculated according to the standard curve method.RESULTS Compared with those in the normal group,the serum contents of saturated fatty acids,such as C6:0,C8:0,C12:0,C13:0,C16:0,C18:0,C20:0,C10:0and C14:0,were significantly increased(P<0.05 or P<0.01)in the model group of rats.The contents of unsaturated fatty acids,such as C14:1T,C14:1,C15:1T,C15:1,C16:1T,C16:1,C17:1T,C17:1,C18:1N12T,C18:1N7T,C19:1N12T,C20:1T,C22:1N9T,C22:1N9T,C22:1N9 and C24:1,were significantly increased(P<0.05 or P<0.01).The contents of trans unsaturated fatty acids were significantly increased(P<0.01).Compared with those in the model group,the serum levels of C16:0,C18:0,C20:0,C10:0,C14:0and other SFAs in the intervention group were significantly reduced(P<0.05 or P<0.01).The contents of C14:1T,C14:1,C15:1,C16:1T,C16:1,C17:1,C18:1N12T,C18:1N9T,C18:1N7T,C20:1T and other MUFA were significantly reduced(P<0.05 or P<0.01).TRNFA content was significantly reduced(P<0.05).CON-CLUSION There is a fatty acid metabolism disorder in serum of rats with ischemic stroke,especially the contents of saturated fatty acids,monounsaturated fatty acids and trans-unsaturated fatty acids increase significantly.The intervention of Sanhua Decoction significantly improves the abnormal changes in serum contents of saturated fatty acid,monounsaturated fatty acid and trans unsaturated fatty acid,which regulates the disorder tends of fatty acid profile to be normal.
