黑果枸杞微卫星DNA与花青素、多酚和黄酮的关联分析 被引量:2

摘要 【目的】筛选试验所用的23个微卫星位点中与花青素、多酚、黄酮有关联效应的位点,加快黑果枸杞良种选育进程.【方法】对选取的23个微卫星标记,应用PCR扩增和非变性聚丙烯酰胺凝胶电泳技术检测群体的遗传多样性,并对各位点不同基因型与经济性状进行关联分析.【结果】该群体遗传参数中平均多态信息含量为0.5851,期望杂合度在0.49~0.75之间.关联分析发现:位点c88061;1;1与花青素和黄酮相关;c60533;1;1、KY284857与多酚相关;c58466;1;1与花青素相关;位点KY284852与花青素和多酚相关.【结论】该群体遗传多样性处于较高水平,并且有较强的遗传潜力.在23个微卫星标记中有5个与黑果枸杞中花青素、多酚和黄酮有关联.选出的分子标记可为黑果枸杞良种选育奠定基础. 【Objective】To speed up the selection and breeding of Lycium ruthenicum varieties,23 microsatellite locus that are associated with anthocyanins,polyphenols and flavonoids were screened in the experiment.【Method】For the 23 selected microsatellite markers,PCR amplification and non-denaturing polyacrylamide gel electrophoresis technology was used to detect the genetic diversity of the population,and the correlation analysis of different genotypes and economic traits at each site were conducted.【Result】The average polymorphic information content in the genetic parameters of the population was 0.5851,and the expected heterozygosity was in the range of 0.49 to 0.75.Correlation analysis showed that locus c88061;1;1 was related to anthocyanins and flavonoid,c60533;1;1 and KY284857 were related to polyphenols,c58466;1;1 was related to anthocyanins,and Locus KY284852 was associated with anthocyanins and polyphenol.【Conclusion】It was indicated that the genetic diversity of this population was at a high level and had strong genetic potential.There were 5 locus associated with anthocyanins,polyphenols and flavonoids among the 23 microsatellite markers in L.ruthenicum.The selected molecular markers can lay a foundation for the selection and breeding of L.ruthenicum varieties.
