肉苁蓉饮片及相关产品中苯乙醇苷的鉴别及测定 被引量:3

摘要 目的:建立肉苁蓉市售饮片及相关产品(提取物、保健品)中4种苯乙醇苷(松果菊苷、管花苷A、毛蕊花糖苷、异毛蕊花糖苷)的薄层色谱鉴别及一测多评(QAMS)含量测定的方法。方法:(1)TLC法,以聚酰胺薄膜为薄层板,三氯甲烷-甲醇-冰乙酸-水(1.5∶2∶1∶7)为展开剂,紫外灯365nm下检视,建立肉苁蓉4种苯乙醇苷的薄层鉴别方法。(2)以松果菊苷为内参物,建立该成分与管花苷A、毛蕊花糖苷和异毛蕊花糖苷的相对校正因子,利用相对校正因子计算各成分的含量,同时比较一测多评法计算结果和外标法实测结果,验证所建立方法的可行性和准确性。结果:肉苁蓉饮片、提取物及保健品中4种苯乙醇苷的TLC分离良好,斑点清晰,Rf值适中。管花苷A、毛蕊花糖苷、异毛蕊花糖苷的相对校正因子的重现性良好,一测多评法和外标法含量结果无明显差异。结论:本研究建立的方法简便准确,重复性好,可为肉苁蓉饮片、提取物和保健食品有效成分质量控制提供参考。 Objective:To establish a method for identification and determination of four phenylethanol glycosides(echinacoside,tubuloside A,acteoside,isoacteoside)in cistanche deserticola pieces,its extracts and related healthy foods by TLC and QAMS.Method:(1)TLC:Four phenylethanol glycosides were identified by TLC with polyamide film as the stationary phase and chloroform-methanol-acetic acid-water(1.5∶2∶1∶7)as the developing solvent.The spot was detected under ultraviolet light at 365 nm.(2)Using echinacoside as internal reference,and the relative correction factors for tubuloside A,acteoside,and isoacteoside were used to calculate the content of each component.The calculation results of QAMS method were compared with the values of external standard method.Result:The TLC spots of 4 phenylethanol glycosides(echinacoside,tubuloside A,acteoside,and isoacteoside)in cistanche decoction pieces,extracts,and functional foods were all clear with good separation,strong specific moderate Rf value.The reproducibility of tubuloside A,acteoside,and isoacteoside relative correction factors were good,and no obvious difference between the two methods.Conclusion:The method established in this study is simple,accurate,and reproducible.It can provide a reference for the quality control of effective components of cistanche decoction pieces,extracts and functional foods.
