跨膜丝氨酸蛋白酶4在乳腺癌细胞MCF-7中影响细胞侵袭和转移的作用机制 被引量:1

摘要 目的研究跨膜丝氨酸蛋白酶4(TMPRSS4)在乳腺癌的发生和发展,以及侵袭和转移中所发挥的生物学作用,旨在为乳腺癌的诊断和治疗找到新的靶点和理论依据。方法采用实时荧光定量聚合酶链反应(qPCR)和Western blot检测乳腺癌细胞中TMPRSS4的mRNA和蛋白表达水平;通过慢病毒转染乳腺癌细胞,干涉TMPRSS4表达水平,采用qPCR和Western blot检测干涉效果;采用细胞划痕实验和Transwell小室实验检测乳腺癌细胞转染前后细胞转移和侵袭能力的改变情况。结果TMPRSS4在多种乳腺癌细胞中的mRNA和蛋白水平均呈高表达;慢病毒转染后,TMPRSS4在MCF-7细胞中的mRNA和蛋白表达水平均明显下降;且慢病毒转染后,MCF-7细胞的转移和侵袭能力与未转染MCF-7细胞和不含干涉片段的病毒转染MCF-7细胞比较均明显减弱。结论TMPRSS4在乳腺癌细胞中呈高表达,并且能够增强乳腺癌MCF-7细胞的转移和侵袭能力。 Objective To study the biological role of transmembrane serine protease 4(TMPRSS4)in the occurrence and development of breast cancer,as well as invasion and metastasis,in order to find new targets and theoretical basis for the diagnosis and treatment of breast cancer.Methods Real-time quantitative polymerase chain reaction(qPCR)and Western blot were used to detect the mRNA and protein expression levels of TMPRSS4 in breast cancer cell.Breast cancer cell were transfected with lentivirus to interfere with the expression level of TMPRSS4,and qPCR and Western blot were used to detect the interference effect.The changes of cell metastasis and invasion ability of breast cancer cells before and after transfection were detected by cell scratch assay and Transwell chamber assay.Results TMPRSS4 mRNA and protein levels were highly expressed in various breast cancer cells.After lentiviral transfection,the mRNA and protein expression levels of TMPRSS4 were significantly decreased in MCF-7 cells.And after lentivirus transfection,the metastasis and invasion ability of MCF-7 cells were significantly weakened compared with untransfected MCF-7 cell and virus-transfected MCF-7 cell without interference fragments.Conclusion TMPRSS4 is highly expressed in breast cancer cell and could promote the metastasis and invasion ability of breast cancer MCF-7 cells.
