摘要
优化草珊瑚提取条件,建立测定草珊瑚中绿原酸、异嗪皮啶、迷迭香酸3种有效成分含量的高效液相色谱分析方法。采用Agilent InfinityLab Poroshell 120 EC-C_(18)色谱柱(150 mm×4.6 mm,4μm),以乙腈/0.1%磷酸水溶液为流动相,梯度洗脱,流速0.6 mL/min,柱温40℃,吸收波长344 nm。结果表明:绿原酸、异嗪皮啶、迷迭香酸浓度分别在0.50~20.0、0.5~20、5.0~100.0 ug/mL范围内与峰面积呈良好的线性关系。其检测限分别为:0.025、0.010、0.10 mg/kg,日内与日间回收率(n=6)在91.8%~108.3%范围内,相对标准偏差在1.5%~4.8%之间。该方法简单高效、准确,灵敏度高且重复性好,适用于草珊瑚中绿原酸、异嗪皮啶、迷迭香酸的含量测定。
The extraction conditions of Sarcandra glabra were optimized,and an HPLC method for the determination of chlorogenic acid,isozinpidine and rosmarinic acid in Sarcandra glabra was established.Agilent infinitylab poroshell 120 EC-C_(18) column(150 mm×4.6 mm,4μm)was used,with acetonitrile-0.1%phosphoric acid aqueous solution as the mobile phase,gradient elution,flow rate of 0.6 ml/min,column temperature of 40℃and absorption wavelength of 344 nm.The results showed that the concentrations of chlorogenic acid,isozinpidine and rosmarinic acid had a good linear relationship with the peak area in the range of 0.50~20.0 ug/ml,0.5~20 ug/ml and 5.0~100.0 ug/ml,respectively.The detection limits were 0.025 mg/kg,0.010 mg/kg and 0.10 mg/kg respectively.The intra day and inter day recoveries(n=6)were in the range of 91.8~108.3%,and the relative standard deviation was in the range of 1.5~4.8%.The method was simple,efficient,accurate,sensitive and reproducible.It was suitable for the determination of chlorogenic acid,isozinpidine and rosmarinic acid in grass coral.
