摘要
目的探究溃疡性结肠炎(ulcerative colitis,UC)患者结肠黏膜组织中黏液基因蛋白2(mucin2,MUC2)、表皮生长因子受体(epidermal growth factor receptor,EGFR)、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达,并分析其与UC病情严重程度、内镜下病变严重程度的相关性。方法选取2017年6月~2020年10月潍坊市人民医院收治的54例UC患者(轻度13例,中度32例,重度9例),另选取同期10例健康对照者的结肠镜活检标本,采用免疫组化法检测结肠黏膜组织中MUC2、EGFR、PCNA表达,比较两组MUC2、EGFR、PCNA表达水平之间的差异,分析黏膜组织中MUC2、EGFR、PCNA水平与UC不同病情严重程度及Mayo内镜下评分(mayo endoscopic subscore,MSE)的相关性,并分析EGFR及MUC2与PCNA之间的相关性。结果UC组的EGFR、MUC2表达水平低于对照组(P<0.05),PCNA表达水平高于对照组(P<0.05)。不同疾病严重程度及MSE的UC患者EGFR、MUC2、PCNA表达水平比较,差异有统计学意义(P<0.05)。EGFR、MUC2表达水平与UC患者的疾病严重程度及MSE呈负相关,而PCNA表达水平与疾病严重程度及MSE呈正相关。结论UC患者结肠黏膜组织中MUC2、EGFR表达水平降低,PCNA表达水平升高,与患者病情严重程度有关,可能在UC发病及病情发展过程中起重要作用。
Objective To investigate the expression of mucin2(Muc2),epidermal growth factor receptor(EGFR)and proliferating cell nuclear antigen(PCNA)in the colonic mucosa of patients with ulcerative colitis(UC).Proliferating cell nuclear antigen(PCNA)expression and correlation with severity of UC disease and endoscopic lesion severity were analyzed.Methods Fifty-four UC patients(13 mild,32 moderate and 9 severe)admitted to Weifang People′s Hospital from June 2017 to October 2020 were selected,and colonoscopic biopsy specimens from 10 healthy controls at the same period were selected.The differences between the expression levels of MUC2,EGFR and PCNA in the mucosal tissues of the two groups were compared,and the correlation between the levels of MUC2,EGFR and PCNA in the mucosal tissues and the different severity of UC and the Mayo endoscopic subscore(mayo endoscopic subscore,MSE)was analysed,as well as the correlation between EGFR and MUC2 and PCNA.Results The expression levels of EGFR and MUC2 in the UC group were lower than those in the control group(P<0.05),and the expression levels of PCNA were higher than those in the control group(P<0.05);the differences between the expression levels of EGFR,MUC2 and PCNA in UC patients with different disease severity and MSE were statistically significant(P<0.05).The expression levels of EGFR and MUC2 were negatively correlated with the disease severity and MSE of UC patients,while the expression levels of PCNA were positively correlated with the disease severity and MSE.Conclusion The decreased expression levels of MUC2 and EGFR and increased expression levels of PCNA in colonic mucosa tissues of patients with UC are related to the severity of the disease and may play an important role in the pathogenesis and progression of the disease in UC.
