摘要
骨质疏松症已经成为与衰老相关的最重要的疾病之一。女性发生率高的原因是由于绝经后雌激素水平的降低伴有骨吸收的增加。最新的研究显示,免疫系统可能是参与绝经后骨质疏松的重要因素,作用于成骨细胞及破骨细胞的促炎因子,以及免疫系统的激活被认为是引起骨质疏松的危险因素,其中白介素-1(IL-1)作为强有力的破骨细胞刺激因子,在生理状态下以及炎症性疾病、绝经后全身性骨量丢失中均发挥着重要作用。本文就IL-1家族、IL-1在细胞、信号通路、动物模型等方面的文献进行回顾,并就IL-1抑制剂、拮抗剂及其衍生物对骨代谢作用进行综述。
Osteoporosis has become one of the most important diseases related to aging.The high incidence in women is due to the decrease of estrogen level and the increase of bone resorption after menopause.The latest research shows that the immune system may be an important factor involved in postmenopausal osteoporosis.The pro-inflammatory factors acting on osteoblasts and osteoclasts,as well as the activation of the immune system,are considered to be risk factors for osteoporosis.Interleukin-1(IL-1),as a powerful osteoclast stimulating factor,plays an important role in the physiological state,inflammatory diseases,and postmenopausal systemic bone loss.In this paper,we review the literature about IL-1 family,the role of IL-1 in different levels including cellular,signaling pathways,and animal models,as well as the effects of IL-1 inhibitors,antagonists,and their derivatives on bone metabolism.
