春尺蠖嗅觉受体基因的鉴定与差异分析 被引量:1

摘要 为鉴定获取春尺蠖嗅觉受体基因,初步探究雌雄差异表达受体基因在对气味分子识别中的作用,基于本实验室已测春尺蠖雌雄成虫转录组数据,共筛选鉴定13个嗅觉受体(odorant receptor,OR)蛋白基因,分别命名为AcinOR1~AcinOR13,并对其信号肽、分子量、等电点进行了预测,同时进行了序列对比与进化关系分析,初步分析雌雄成虫中13个嗅觉受体蛋白基因的差异表达情况。结果发现,AcinOR1~AcinOR13长度在68~415 aa之间,分子量在7.91~47.32 kDa之间,等电点在4.42~10.03之间,均未发现信号肽。系统进化分析发现,AcinOR2/AcinOR5/AcinOR13、AcinOR13/AcinOR7分别聚在一支上,同时AcinOR1/BmorOR10、AcinOR4/HarmOrco、AcinOR10/EoblOR4、AcinOR3/EoblOR29和AcinOR8/ScinOR5聚在一支上;此外,表达分析结果发现,AcinOR6和AcinOR9呈雌性高表达,AcinOR1呈雄性高表达,其他基因在雌雄个体间表达无显著差异。本研究为探究春尺蠖的嗅觉受体基因的功能奠定了基础,同时为该虫生物防治提供新思路。 In order to identify and acquire olfactory receptor genes of Apocheima cinerarius,the role of male and female differentially expression receptor genes in odor molecular recognition was preliminarily investigated. Based on transcriptome data of male and female adults of A. cinerarius,13 odorant receptors(OR)protein genes,named AcinOR1-AcinOR13 were screened and identified and the signal peptides,molecular weights and isoelectric points of target protein genes were predicted. Meanwhile,sequence comparison and evolutionary relationship analysis were conducted and the differential expression of 13 olfactory receptor protein genes in male and female adults was analyzed. The results showed that the length of AcinOR1-AcinOR13 ranged from 68 to 415 aa and the molecular weight ranged from 7.91 to 47.32 kDa and the isoelectric point ranged from 4.42 to 10.03 and signal peptide was not found. Phylogenetic analysis showed that AcinOR2/AcinOR5/AcinOR13 and AcinOR13/AcinOR7 clustered on the same branch,respectively. Meanwhile,AcinOR1/BmorOR10,AcinOR4/HarmOrco,AcinOR10/EoblOR4,AcinOR3/EoblOR29,and AcinOR8/ScinOR5 were clustered on one branch. In addition,expression analysis showed that AcinOR6 and AcinOR9 were highly expressed in females,while AcinOR1 was highly expressed in males and there was no significant difference in the expression of other genes between male and female individuals. This study laid a foundation for exploring the function of olfactory receptor gene of A. cinerarius,and provided new ideas for biological control of A. cinerarius.
