沉默OGFR基因对非小细胞肺癌A549细胞株增殖、迁移和侵袭能力的影响

摘要 目的探讨沉默OGFR基因对非小细胞肺癌A549细胞株增殖、迁移和侵袭的影响。方法取A549细胞株,分为对照组、实验组,分别转染si-NC阴性对照、OGFR特异性的小干扰RNA。采用RT-qPCR和Westernblotting法分别检测细胞中的OGFRmRNA和蛋白,采用CCK-8实验检测细胞增殖能力,细胞划痕实验检测细胞迁移能力,Transwell侵袭实验检测细胞侵袭能力,平板克隆形成实验检测细胞集落形成能力。结果与对照组相比,实验组OGFRmRNA及蛋白相对表达量低,培养24、48、72 h细胞增殖OD值均低,穿膜细胞数少,细胞迁移率低,克隆形成细胞集落数少(P均<0.05)。结论沉默OGFR基因可降低A549细胞株OGFR基因mRNA和蛋白质的表达,在体外减弱了其增殖、迁移和侵袭能力。 Objective To investigate the effects of silencing OGFR gene on the proliferation,migration and invasion of non-small cell lung cancer A549 cell line.Methods A549 cells were divided into control group and experimental group,and transfected with si-NC negative control and OGFR specific small interfering RNA respectively.RT-qPCR and Western blotting were used to detect OGFR mRNA and protein in cells,CCK-8 method was used to detect cell prolifera⁃tion,cell scratch test was used to detect cell migration,Transwell invasion test was used to detect cell invasion,and plate clone formation was used to detect cell colony formation.Results Compared with the control group,the relative expres⁃sion of OGFR mRNA and protein in the experimental group was low,the proliferation OD value of cells cultured for 24,48 and 72 hours was low,the number of transmembrane cells was small,the cell migration rate was low,the colony number of clonogenic cells was small(P<0.05).Conclusion Silencing OGFR gene can reduce the expression of OGFR gene mRNA and protein in A549 cell line,and weaken its proliferation,migration and invasion in vitro.
