摘要
目的 研究茚三酮显色-可见分光光度法和高效液相色谱法测定大蒜总氨基酸含量结果间的相关性。方法 分别采用茚三酮显色-可见分光光度法和高效液相色谱法测定大蒜总氨基酸含量,对两种测定方法的结果采用数字序列相关性分析法建立相关性数学模型。结果 基于数字序列相关性分析法建立相关性数学模型得到 Cμ_(1)·μ_(2)=0.921 7,r= 0.932 0,说明大蒜总氨基酸的两种含量测定方法具有高度相关性,拟合出线性方程:Y= 1.003 3X+2.278 1,r= 0.932 0,且 P<0.01。3批验证的大蒜总氨基酸含量数据:茚三酮显色-可见分光光度法测定值分别为 13.00%、7.79% 和 8.73%;高效液相色谱法实测值分别为 15.40%、9.98% 和 11.60%;高效液相色谱法测算值分别为 15.32%、10.09% 和11.03%。结论 高效液相色谱法和茚三酮显色-可见分光光度法测定大蒜总氨基酸含量的结果之间具有良好的相关性,通过线性方程可由一种大蒜总氨基酸含量测定结果预测另一方法的测定结果。
Objective To study the correlation between ninhydrin color visible spectrophotometry and HPLC in the determination of total amino acids in garlic.Methods The total amino acid content of garlic was determined by ninhydrin color-visible spectrophotometry and high performance liquid chromatography,respectively.The numerical sequence correlation analysis method was used to establish a correlation mathematical model for results of the two determination methods.Results The correlation mathematical model was established based on the numerical sequence correlation analysis method,and Cμ_(1)·μ_(2)=0.9217,r= 0.9320,indicating that the two methods for determination of total amino acids in garlic were highly correlated,and a linear equation was fitted:Y= 1.0033X +2.2781,r=0.9320,and P<0.01.The data of total amino acid content of garlic verified by three batches:ninhydrin color-visible spectrophotometry measured values were 13.00%,7.79% and 8.73% respectively;the measured values by high performance liquid chromatography were 15.40%,9.98% and 11.60% respectively;the calculated values by high performance liquid chromatography were 15.32%,10.09% and 11.03% respectively.Conclusion There is a good correlation between the results of determination of total amino acid content in garlic by high performance liquid chromatography and ninhydrin color-visible spectrophotometry.
