一株沙福芽胞杆菌生长及其产碳酸酐酶特性

摘要 为了探明一株沙福芽胞杆菌(Bacillus safensis)的生长与产碳酸酐酶(carbonic anhydrase,CA)特性,采用单因素实验,研究了培养基初始pH、接种量、温度、摇床转速及Zn^(2+)、Co^(2+)、Cd^(2+)、Mn^(2+)、Mg^(2+)、Ca^(2+)等金属离子对细菌生长及其产CA活性的影响。结果表明,该菌株的最佳生长条件和产CA的最佳条件不同。30℃时,细菌生长最快,45℃时酶活性最高;细菌进入衰亡期酶活性达到最高。胞内外总CA检测结果表明,该菌株产胞内CA。加入0.010 mmol/L Mn ^(2+),酶活性达2.46 U/mL,比对照组提高了2.6倍。 In order to explore the characteristics on growth and carbonic anhydrase(CA)producing strain of Bacillus safensis,the effects of factors including initial pH,inoculation size,temperature,shaker rotational speed,and metal ions of Zn^(2+),Co^(2+),Cd^(2+),Mn^(2+),Mg^(2+),and Ca^(2+)on the bacterial growth and CA activity were investigated by single factor experiment.The results showed that the optimal conditions for the growth and CA production of this strain were different.The bacteria grew fastest at 30℃,and the enzyme activity was the highest at 45℃.Furthermore,the bacteria had the highest enzyme activity in decline phase.The results from the test of intracellular and extracellular total CA activity suggested that this strain produced intracellular CA.By adding 0.010 mmol/L Mn^(2+),the enzyme activity reached 2.46 U/mL,increased 2.6 times higher as compared with control group.
