摘要
目的:研究巴西菇水提物对高脂膳食大鼠的降脂降糖作用。方法:40只成年SD大鼠随机分为空白对照组、高脂模型组、巴西菇水提物(低、中、高剂量)干预组5组,每组8只。其中,空白对照组给予普通饲料;其余各组给予高脂饲料;干预组分别以10 mg/kg、20 mg/kg、30 mg/kg剂量灌胃,每天1次,连续40 d。实验结束后处死大鼠,分析血糖血脂、胰岛素、抗氧化酶水平。结果:巴西菇水提物组的大鼠血糖、血脂、胰岛素水平较高脂模型组显著降低,体重也明显降低(P<0.05,P<0.01);巴西菇水提物显著降低了高脂大鼠肝脏、肾脏丙二醛(Malondialdehyde,MDA)水平,提高了超氧化物歧化酶(Superoxide Dismutase,SOD)、过氧化氢酶(Catalase,CAT)和谷胱甘肽过氧化物酶(Glutathione Peroxidase,GSH-Px)活性(P<0.05,P<0.01);巴西菇水提物显著降低了高脂大鼠肝脏PPAR-γ的mRNA水平、提高了肾脏UCP-1的mRNA水平。结论:巴西菇水提物可有效改善高脂膳食大鼠的血糖、血脂水平,提高抗氧化能力。这可能与它调节PPAR-γ和UCP-1的mRNA表达水平有关。
Objective:Study on the effects of water extract of Brazilian mushroom on the blood glucose and lipids in rats fed with high fat diet.Methods:Forty adult SD rats were randomly divided into 5 groups:blank control group,high fat model group,and water extract of Brazilian mushroom(low,medium and high doses)intervention groups,with 8 rats in each group.Among them,the blank control group was given normal diet;the other groups were given high fat diet;and the intervention group was gavaged at 10 mg/kg,20 mg/kg and 30 mg/kg,respectively once a day for 40 days.After the end of the experiment,the rats were sacrificed and the levels of blood glucose,lipid,insulin and antioxidant enzymes were analyzed.Results:The levels of blood glucose,lipid and insulin in the water extract of Brazilian mushroom group were significantly decreased with comparison to the high fat model group,and the body weight was also significantly decreased(P<0.05,P<0.01).At the same time,the water extract of Brazilian mushroom significantly decreased the MDA levels in liver and kidney of high fat rats,and increased the activities of SOD,CAT and GSH-Px(P<0.05,P<0.01).The water extract of Brazilian mushroom significantly reduced liver PPAR-γmRNA level,increased the expression of kidney UCP-1 mRNA in rat.Conclusion:The water extract of Brazilian mushroom can effectively improve the blood glucose and lipid levels and enhance the antioxidant capacity of rats fed with high fat diet,which would correlate with the regulation of the expression of PPAR-γand UCP-1 mRNA.
