某中草药颗粒对小鼠辅助免疫调节功能的生理评价

摘要 目的了解某中草药颗粒对小鼠免疫力的辅助调节作用。方法将某中草药颗粒含量设低(1.00 g/kg·bw)、中(2.00 g/kg·bw)、高(6.00 g/kg·bw)剂量组及蒸馏水对照组,给小鼠经口灌胃连续30 d,采用碳廓清实验、ConA诱导的小鼠淋巴细胞转化实验及NK细胞活性测定、脏体比值测定及半数溶血值(HC50)测定和抗体生成细胞数测定、迟发型变态反应实验(DTH)、小鼠腹腔巨噬细胞吞噬鸡红细胞实验的方法检测相应功效学指标,用SPSS 19.0软件进行单因素方差分析及Dunnett检验,以P<0.05为差异有统计学意义。结果本研究实验末,中剂量组小鼠致敏后24 h足跖肿胀度为0.44±0.11 mm、高剂量组小鼠为0.51±0.17 mm,与对照组0.26±0.17 mm比较显著提高(t=2.691,P=0.028;t=3.662,P=0.002);中剂量组小鼠NK细胞活性平方根反正弦转换值为(35.30±6.61)、高剂量组小鼠为(37.32±6.16),与对照组(28.06±6.01)比较显著提高(t=2.737,P=0.026;t=3.490,P=0.04);高剂量组的小鼠抗体生成细胞数为211±59个/106个脾细胞,与对照组126±53个/106个脾细胞相比提高(t=3.054,P=0.012);高剂量组小鼠HC50值为215.14±51.18,较对照组155.25±34.75提高(t=3.033,P=0.012)。各剂量对小鼠体重及增重、胸腺及脾脏与体重的比值、碳廓清能力、淋巴细胞转化能力、巨噬细胞吞噬鸡红细胞能力的影响均无统计学意义(P>0.05)。结论本研究中草药颗粒对实验小鼠具有辅助调节免疫力功能。 Objective To investigate the auxiliary regulation effect of a Chinese herbal medicine granule on immunity in mice.Methods Low(1.00 g/kg·BW),medium(2.00 g/kg·BW),and high(6.00 g/kg·BW)dose groups and the control group(distilled water)were set.Mice in each of those groups were gavaged for 30 consecutive days.The effects of the Chinese herbal medicine granules on the immune function of mice were evaluated by carbon clearance experiment,lymphocyte transformation induced by ConA,NK cell activity,visceral body ratio,Half hemolysis value(HC50),antibody-producing cell count,delayed type allergy test,and abdominal macrophage phagocytosis of chicken erythrocytes.The data were analyzed by one-way ANOVA,followed by Dunnett tests in SPSS19.0 software.P<0.05 was considered as statistically significant.Results At the end of this study,the plantar swelling of mice in the medium-dose group(0.44±0.11 mm)and high-dose group(0.51±0.17 mm)were significantly higher than that in the control group(0.26±0.17 mm,t=2.691,P=0.028;t=3.662,P=0.002).The square root inverse sinusoidal conversion value of NK cell activity in medium-dose group and high-dose group was significantly higher than that in control group(t=2.737,P=0.026;t=3.490,P=0.04).The number of antibody generating cells in high-dose group was significantly higher than that in the control group(211±59/106 vs.126±53/106 splenocytes,t=3.054,P=0.012).The HC50 value of high-dose group was significantly higher than that of the control group(215.14±51.18 vs.155.25±34.75;t=3.033,P=0.012).There were no significant effects of the Chinese herbal medicine granules on the body weight and weight gain,the ratio of thymus and spleen to body weight,carbon clearance ability of mononuclear macrophages,ability of lymphocyte transformation and macrophages to phagocyte chicken erythrocytes of mice(P>0.05).Conclusions In this study,the Chinese herbal medicine granules had the function of auxiliary regulating immunity in experimental mice.
