盘羊与欧拉羊杂交后代UCP2、UCP3基因CDS区克隆与生物信息学分析

摘要 研究旨在通过对盘羊与欧拉羊杂交后代UCP2、UCP3基因CDS区克隆并进行生物信息学分析,了解其生物学特性和结构。结果表明,杂交后代羊UCP2基因CDS区长931 bp,共有7个开放阅读框,编码309个氨基酸;UCP3基因CDS区长937 bp,共有5个开放阅读框,编码244个氨基酸。杂交后代羊与绵羊的亲缘关系最为相近,与马和西伯利亚雪橇犬的亲缘关系最远。杂交后代羊UCP2理论等电点9.64,不稳定指数36.27,半衰期30 h,总平均亲水性0.109,为稳定蛋白且表现疏水性;杂交后代羊UCP3理论等电点9.25,不稳定指数44.80,半衰期30 h,总平均亲水性-0.034,为不稳定蛋白且表现亲水性。UCP2、UCP3均属于非分泌蛋白且无信号肽。UCP2、UCP3都不具有跨膜螺旋结构;UCP2存在26处磷酸化修饰位点,UCP3存在23处磷酸化修饰位点。UCP2、UCP3蛋白二级结构组成均以α-螺旋为主。同时,通过亚细胞定位发现UCP2、UCP3蛋白均主要存在于细胞质内。研究结果对今后探索UCP2、UCP3生理功能可提供一定的科学基础和依据。 This study aims to clone and do bioinformatics analysis on the CDS region of UCP2 and UCP3 genes in Argali and Oula Tibetan sheep hybrid offspring,for understanding its biological characteristics and structure.The CDS region of UCP2 gene in crossbred sheep was 931 bp,with 7 open reading frames,encoding 309 amino acids.The CDS region of UCP3 gene was 937 bp,with 5 open reading frames,encoding 244 amino acids.The hybrids are most closely related to sheep and most distantly related to the horse and the siberian husky.UCP2 is a stable protein with a theoretical isoelectric point of 9.64,instability index of 36.27,half-life of 30 h and total average hydrophilicity of 0.109 and performance hydrophobicity.UCP3 is an unstable protein with a theoretical isoelectric point of 9.25,an instability index of 44.80,half-life of 30 h,and a total average hydrophilicity of-0.034 and show hydrophilicity.UCP2 and UCP3 are both non-secretory proteins and have no signal peptides.UCP2 and UCP3 have no transmembrane helical structure,there were 26 phosphorylation sites in UCP2 and 23 phosphorylation sites in UCP3.The secondary structure of UCP2 and UCP3 proteins mainly consists ofα-helix.At the same time,the subcellular locali-zation revealed that UCP2 and UCP3 proteins mainly existed in the cytoplasm.This study can provide a scientific basis for exploring the physiological functions of UCP2 and UCP3 in the future.
