摘要
目的了解1例从广西口岸入境的澳门手足口病病例的病毒基因组序列特征。方法提取待测鼻咽拭子样本的病毒RNA,采用实时荧光RT-PCR方法检测肠道病毒核酸,同时将扩增产物进行测序,通过MEGA 7.0软件使用邻位相接法构建进化树分析。结果本次澳门手足口病病例为柯萨奇病毒A组6型,与CV-A6的Gdula原型株的核苷酸同源性为81%。基因型为D3a分支,与2018年的济南株(MK357084、MK357085)、2017年的广西株(MK839021)、2017年的青岛株(MN689954)具有较高的遗传同源性。结论本研究提示,目前澳门手足口病可能以CV-A6的D3a进化分支为流行株。
Objective To understand the characteristics of the viral genome sequence of a case of hand,foot,and mouth disease(HFMD)entering the Guangxi port from Macao.Methods The viral RNA was extracted from nasopharyngeal swabs,enterovirus nucleic acid was detected by fluorescent Real-time RT-PCR.At the same time,the amplified products were sequenced.A phylogenetic tree was constructed by MEGA 7.0 software with adjacent-position method.Results The HFMD case came from Macao was Coxsackie virus group A type 6,and its nucleotide homology with the Gdula prototype strain of CV-A6 was 81%.Its genotype was a branch of D3a,which had high genetic homology with the Jinan strain in 2018(MK357084,MK357085),the Guangxi strain in 2017(MK839021),and the Qingdao strain in 2017(MN689954).Conclusion This study suggests that the D3a evolutionary branch of CV-A6 may be the prevalent strain of hand,foot,and mouth disease in Macao.
