黄金万红膏对银屑病模型小鼠治疗效果研究 被引量:3

摘要 探讨黄金万红膏对咪喹莫特诱导银屑病小鼠的治疗效果.首先将60只6~8周,雌雄各半的Babl/c小鼠,背部脱毛形成2 cm×3 cm的造模区域,随机分为空白对照组、咪喹莫特组、黄金万红膏高剂量组(200 mg/d)、万红膏中剂量组(100 mg/d)、黄金万红膏低剂量组(50 mg/d)、卡泊三醇组(200 mg/d),造模及治疗结束后,根据小鼠银屑病严重程度评分标准观察小鼠银屑病皮损变化,病理切片法观察皮损组织形态学变化.用q-PCR技术检测小鼠背部皮肤组织中IL-17A、IL-17F、IL-23 mRNA的表达量,利用ELISA检测小鼠血清中IL-17、TNF-α、IL-22的表达量.与模型组相比,万红膏治疗组的银屑病样皮损减少,严重程度评分(PASI)降低.组织中IL-17A、IL-17F、IL-23的mRNA量及血清中IL-17、TNF-α、IL-22均有减少.研究表明万红膏可通过调节IL-17A、IL-17F、IL-22、IL-23、TNF-α来减轻银屑病的症状,并且其作用效果明显比阳性药卡泊三醇更为温和. his study is to investigate the therapeutic effect of HuangjinWanhong ointment on imiquimod-induced psoriasis mice.Sixty 6-week-old Babl/c mice in both sexes were depilated on the back to form a 2 cm×3 cm modeling area,and randomly divided into blank control group,imiquimod group,HuangjinWanhong ointment high-dose group(200 mg/d),HuangjinWanhong ointment medium-dose group(100 mg/d),Huangjin Wanhong ointment low-dose group(50 mg/d),and carbotriol group(200 mg/d).After modeling and treatment,the changes of psoriasis lesions in mice were observed according to the criteria for scoring the severity of psoriasis,and the histomorphological changes of lesions were observed by pathological sectioning.The expressions of IL-17A,IL-17F and IL-23 mRNA in the skin tissue of mouse back were detected by q-PCR,and the expressions of IL-17,TNF-αand IL-22 in the serum of mice were detected by ELISA.Compared with the model group,the psoriasis-like lesions in the HuangjinWanhong ointment-treated group were reduced,the severity score(PASI)was decreased,and the amount of mRNA of IL-17A,IL-17F and IL-23 in tissues and IL-17,TNF-αand IL-22 in serum were reduced.This study showed that HuangjinWanhong ointment can reduce the symptoms of psoriasis by regulating IL-17A,IL-17F,IL-22,IL-23,and TNF-α,and its effect is obviously milder than the positive drug calcipotriol.
