摘要
目的探究野菊花对大脑中动脉闭塞诱导的缺血性脑损伤的保护作用及相关神经功能的影响。方法使用线栓法建立大脑中动脉闭塞模型(MCAO),将成年雄性昆明小鼠随机分为假手术(Sham)组、缺血/再灌注(I/R)组、溶剂对照(S+P)组和野菊花(FCI)组。采用氯化三苯基四氮唑(TTC)染色法检测小鼠脑梗死面积;使用行为学实验检测缺血对小鼠神经功能的影响;采用TUNEL染色法检测大脑缺血区细胞凋亡情况。结果与I/R组相比,FCI组小鼠脑梗死面积明显减小(P<0.05),缺血区细胞凋亡数目明显减少(P<0.05);在Morris水迷宫实验的定位航行实验中,与I/R组相比,FCI组寻找到平台的潜伏期显著缩短(P<0.05),空间探索实验中,FCI组在目标象限的停留时间显著延长(P<0.05);在关联恐惧实验中,与I/R组相比,FCI组凝滞时间显著增加(P<0.05)。结论野菊花对小鼠大脑中动脉闭塞导致的缺血性脑损伤具有明显的保护作用,其作用有可能是通过抑制小鼠脑缺血区神经细胞凋亡来完成的。
Objective To explore the protective effect of Flos Chrysanthemi indici on ischemic injury induced by middle cerebral artery occlusion and its influence on related neurological functions.Methods A mouse model of middle cerebral artery occlusion(MCAO)was established using the suturing method.Adult male Kunming mice were randomly divided into four groups:a Sham group,an ischemia/reperfusion(I/R)group,and a solvent control(S+P)group and a Flos Chrysanthemi indici injection(FCI)group.The cerebral infarction size was measured by triphenyltetrazolium chloride(TTC)staining method.The effect of ischemia on the neurological function of mice were detected by behavioral experiments.Cell apoptosis in cerebral ischemic areas were examined by TUNEL assay.Results Compared with the I/R group,the FCI group showed remarkably reduced cerebral infarction size(P<0.05),and a significantly decreased number of apoptotic cells in the ischemic area(P<0.05).In the Morris water maze,compared with the I/R group,the FCI group presented a significantly shortened latency to find a platform during navigation(P<0.05).According to the spatial probe test,the FCI group showed a significantly increased number of crossings of the hidden platform,compared with the I/R group.In the fear conditioning test,compared with the I/R group,the freezing time of the FCI group significantly increased(P<0.05).Conclusions Flos Chrysanthemi indici exhibits obvious protective effects on ischemic injury induced by middle cerebral artery occlusion in mice,which may be related to inhibiting cell apoptosis in the ischemic area of mice.
