柯里拉京对照品的制备及其结构鉴定

Preparation and structure identification of corilagin reference substance

目的制备柯里拉京对照品并鉴定其结构。方法采用叶下珠属植物叶下珠,加水煎煮浸提,滤过,滤液分别通过D101大孔吸附树脂、Diaion HP大孔吸附树脂、葡聚糖凝胶(Sephadex LH-20)吸附洗脱,收集洗脱液,减压浓缩,加入乙腈(50%~60%)反复重结晶得到类白色结晶。经过纯度分析、含量分析及波谱分析对该结晶进行确证。结果本工艺提取收率可达0.8%。制备所得柯里拉京产品纯度高(99.9%以上),且易于实现产业化放大。结论该物质符合中药化学检验用对照品的相关要求,可作为含柯里拉京成分的药材及其制剂质量控制以及中药药效物质基础研究用的化学对照品。

Objective To determine the structure of corilagin reference substance and prepare it.Methods The Phyllanthus urinaria L.was decocted,extracted and filtered with water.The filtrate was eluted by D101 macroporous adsorption resin,Diaion HP macroporous resin and Sephadex LH-20,respectively.The eluent was collected,concentrated and recovered the solvent under reduced pressure before adding acetonitrile(50%~60%)and recrystallizing repeatedly to obtain nearly white crystals.Results The extraction yield of corilagin was 0.8% with high purity(over 99.0%),which was suitable for industrial scale-up.Conclusion The substance meets the requirements of reference substance for chemical inspection of traditional Chinese medicine,and can be used as the chemical reference for the quality control of medicinal materials,preparations containing corilagin,and the basic research of pharmacodynamic substances in traditional Chinese medicine.
