摘要
骨关节炎(OA)是一种以关节软骨退变、滑膜增生及软骨下骨改变为特征的退行性疾病,是造成老年人慢性疼痛最常见的疾病之一。但是,由于OA发病机制没有完全阐明,目前缺乏有效药物阻止疾病进展。近年来,单细胞转录组测序(scRNA-seq)技术可以根据细胞的异质性将某一组织分成不同的细胞亚群,进而对不同的细胞亚群进行独立分析并揭示细胞亚群间相互作用与OA的关系,为深入研究OA的发病机制开辟了新的途径。本文就scRNA-seq的原理,各技术之间的比较,以及scRNA-seq在OA中的应用进行综述,为进一步探索OA的发病机制提供参考。
Osteoarthritis(OA) is a degenerative disease, which is characterized by the degeneration of the articular cartilage, synovial hyperplasia and the changes in the subchondral bone, and is also one of the most common diseases causing chronic pain in the elderly.However, there is no effective drug to prevent the development so far because of the unknown pathogenesis of OA. In recent years, the application of single-cell RNA sequencing(sc RNA-seq) has provided us with a new method to explore the pathogenesis of OA, which can divide the tissue into different cell subsets regarding their heterogeneity and reveal the interaction between cell subsets by performing analysis independently. In order to explore the pathogenesis of OA,we conducted a systematic review for the principle of sc RNAseq, the comparison between different techniques and the application of sc RNA-seq in OA.
