基于GEO和TCGA数据库筛选子宫内膜癌发病及预后的相关基因

摘要 目的通过生物信息学方法分析与子宫内膜癌(EC)发病及预后相关的基因。方法下载基因表达综合数据库(GEO)中相关样本的基因芯片数据,筛选出EC与正常子宫内膜组织细胞中的差异表达基因(DEGs)。运用STRING在线数据库及DAVID在线数据库对DEGs进行蛋白-蛋白互作网络(PPI)分析、基因本体论(GO)及京都基因与基因组百科全书(KEGG)代谢通路分析,并利用Cytoscape软件筛选出关键基因。通过癌症基因组图谱(TCGA)数据库中的子宫内膜癌的生存资料对关键基因进行生存分析。结果本研究共筛选出EC相关108个DEGs,其中10个关键基因为JUN、UBE2I、WT1、GATA2、WNT2、CXCL12、ZEB1、TCF4、FOXL2、PRKCA,均在EC中相对高表达。WNT2、TCF4和FOXL2关键基因对患者的生存率差异有统计学意义(P<0.05)。Wnt信号通路可能对EC的发病有重要作用。结论WNT2、TCF4和FOXL2与EC的发病及预后有着密切关系,为EC新的诊断标志物和治疗靶点筛选提供新思路。 Objective To analyze the genes associated with the onset and prognosis of endometrial cancer(EC)by bioinformatics.Methods The differentially expressed genes(DEGs)in EC and normal endometrial tissues were screened by downloading the gene chip data of Gene Expression Omnibus(GEO).The protein-protein interaction network(PPI)analysis,Gene Ontology(GO)enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)metabolic pathway analysis were performed on DEGs using STRING online database and DAVID online database.Cytoscape software was used to screen out key genes.The survival data of EC in the Cancer Genome Atlas(TCGA)database were used to analyze the survival of key genes.Results A total of 108 DEGs related to EC were screened out in this study,among which 10 key genes were JUN,UBE2I,WT1,GATA2,WNT2,CXCL12,ZEB1,TCF4,FOXL2 and PRKCA,all of which were relatively highly expressed in EC.Key genes WNT2,TCF4 and FOXL2 had significant effects on survival(P<0.05).Wnt signaling pathway may play an important role in the pathogenesis of EC.Conclusion WNT2,TCF4 and FOXL2 are closely related to the pathogenesis and prognosis of EC,providing new ideas for screening new diagnostic markers and therapeutic targets of EC.
