摘要
NR4A1作为孤核受体(NR4A)超家族的成员之一,因其缺乏内源性配体结合位点而与经典的核受体结构不同。特殊的结构使它成为一种可以在多种刺激下快速诱导的即时早期反应基因。目前研究表明,NR4A1是一种转录调控因子,对机体的炎症反应、免疫功能、细胞增殖与分化、代谢等起着转录调控作用。糖尿病的患病率逐年上升,NR4A1与糖尿病之间的相关性研究也倍受关注。近期研究表明,NR4A1的过表达促进胰岛素的分泌和抑制脂肪细胞的分化与成熟,从而发挥抗脂肪作用,进而影响糖脂代谢;同时,NR4A1的缺失也可以增加糖尿病并发症(糖尿病肾病、糖尿病视网膜病变)的易感性。本文对NR4A1与糖尿病之间的相关性作一综述,为以后NR4A1可能作为潜在靶点治疗糖尿病提供科学依据。
As a member of the orphan nuclear receptor(NR4A)superfamily,NR4A1 is different from the classical nuclear receptor structure due to its lack of endogenous ligand binding sites.The special structure makes it an immediate early response gene that can be quickly induced by a variety of stimuli.Current studies have shown that NR4A1 is a transcriptional regulatory factor,which plays a key role in the transcriptional regulatory of inflammatory response,immune function,cell proliferation and differentiation,and metabolism.The prevalence of diabetes mellitus has increased year by year,and the correlation between NR4A1 and diabetes mellitus has also attracted much attention.Recent studies have shown that the overexpression of NR4A1 exerts anti-diabetic effect by promoting the secretion of insulin and inhibiting the differentiation and maturation of adipocytes,thereby affecting glucose and lipid metabolism.The lack of NR4A1 can also increase the susceptibility of diabetic complications such as diabetic nephropathy and diabetic retinopathy.This article reviews the correlation between NR4A1 and diabetes mellitus,and provides a scientific basis for NR4A1 as a potential target for the treatment of diabetes.
