摘要 目的:研究艾绒饮片的薄层鉴别和含量测定方法,提高其质量控制标准。方法:以艾叶为对照药材,以石油醚(60~90℃)-甲苯-丙酮(10∶8∶0.5)为展开剂,2%香草醛硫酸为显色剂,建立艾绒的薄层色谱(TLC)鉴别方法;建立高效液相色谱法(HPLC)测定异泽兰黄素的含量,以CAPCELL PAK C18MGⅡS5(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸溶液(36∶64),流速为1.0 mL/min,检测波长为344 nm,柱温为30℃。结果:建立的TLC鉴别方法,斑点清晰,分离效果较好;HPLC法测定异泽兰黄素的进样量在0.0187~0.561μg(r=0.9999)范围内与峰面积呈现良好的线性关系;精密度、稳定性(24 h)、重复性试验的RSD均小于2.0%;平均回收率(n=6)为97.98%(RSD=1.11%)。结论:建立的方法简便易行,专属性强,重复性好,可用于艾绒饮片的质量控制及评价。 Objective:To improve the quality standards for moxa floss by TLC and quantitate determine.Methods:The TLC method of moxa floss were based on Artemisiae argyi Folium as the control herb,petroleum ether(60-90℃)-tolueneacetone(10∶8∶0.5)as the unfolding agent,and 2%vanillin sulfate was used as colorimetric agent.The contents of epuatilinin was determined by HPLC.The CAPCELL PAK C18MGⅡS5(4.6 mm×250 mm,5μm)was used with the mobile phase of acetonitrile-0.1%phosphoric acid solution(36∶64),at the flow rate of 1.0 mL/min,the detection wavelength was 344 nm,and the column temperature was at 30℃.Results:The spots in the TLC were clear and well-separated.The linear ranges of epuatilinin was 0.0187-0.561μg(r=0.9999),respectively.RSDs of precision,stability(24 h)and repeatability tests were less than 2.0%.The average recoveries(n=6)wase 97.98%(RSD=1.11%).Conclusion:The method is easy,simple,specific and repeatable,which can be used for the quality evaluation and control of Moxa floss.
