摘要
目的 研究琥珀消石颗粒对肾脏草酸钙结石大鼠骨形成蛋白2 (BMP2)通路及炎症应激反应的影响。方法 选择成年雄性SD大鼠作为对照组(n=10),选择成年遗传性高钙尿结石雄性大鼠随机分为模型组(n=10)和治疗组(n=10),分别给予纯水灌胃和琥珀消石颗粒灌胃、连续4周。检测24 h大鼠的尿草酸及钙含量,肾脏内钙盐沉积,肾脏中骨形态发生蛋白2 (BMP2)、Runt相关转录因子2 (RUNX2)表达水平及白介素(IL)-1β、IL-6、肿瘤坏死因子-α (TNF-α)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量。结果 模型组大鼠肾间质内可见明显钙盐沉积,24 h尿草酸含量、钙含量及肾脏中BMP2、RUNX2的表达水平、IL-1β、IL-6、TNF-α、MDA的含量明显高于对照组,SOD含量明显低于对照组,差异均有统计学意义(P<0.05);治疗组大鼠肾间质内钙盐沉积明显减少,24 h尿草酸含量、钙含量及肾脏中BMP2、RUNX2的表达水平、IL-1β、IL-6、TNF-α、MDA的含量明显低于模型组,SOD含量明显高于模型组,差异均有统计学意义(P<0.05)。结论 琥珀消石颗粒能明显改善肾脏草酸钙结石大鼠的肾间质钙盐沉积,这一作用与抑制BMP2通路及炎症应激反应激活有关。
Objective To study the effect of HupoXiaoshi Granule on bone morphogenetic protein 2(BMP2)pathway and inflammatory stress response of rats with renal calcium oxalate stone. Methods Adult male SD rats were selected as control group(n=10). Male rats with hereditary hypercalciuria stone were randomly divided into model group(n=10) and treatment group(n=10), which were given pure water and HupoXiaoshi granules for 4 weeks. The levels of urinary oxalic acid and calcium, calcium deposition in kidney, the expression level of BMP2 and Runx2, and the contents of interleukin-1β(IL-1β), IL-6, tumor necrosis factor-α(TNF-α), malondialdehyde(MDA), and superoxide dismutase(SOD) in kidney were measured. Results In the model group, calcium deposition was observed in the renal interstitium;the contents of oxalic acid, calcium in 24 h, the expression levels of BMP2, RUNX2, and the contents of IL-1β,IL-6, TNF-α, MDA in kidney of model group were higher than those of control group;SOD content was lower than that of control group. Compared with the model group, calcium deposition in renal interstitium of treatment group significantly decreased;the contents of oxalic acid, calcium in 24 h, the expression levels of BMP2, RUNX2, and the contents of IL-1β, IL-6, TNF-α, MDA in kidney of treatment group significantly reduced;SOD content significantly increased;the differences were statistically significant(P<0.05). Conclusion HupoXiaoshi granule can improve the calcium deposition in renal interstitium of rats with renal calcium oxalate stone, which is related to the inhibition of BMP2 pathway and activation of inflammatory stress response.
