摘要
目的:探讨CD44基因沉默对鼻咽癌细胞周期及凋亡的影响。方法:设计及合成CD44 siRNA,转染人鼻咽癌SUNE-15-8F细胞,Real-time qPCR及Western blot评价转染效果,流式细胞仪分析各组细胞周期和凋亡情况。结果:CD44 siRNA显著下调了人鼻咽癌SUNE-15-8F细胞中CD44 mRNA与蛋白表达(P<0.05)。流式细胞仪检测结果显示siRNA组细胞中G0/G1期细胞比例明显升高,S期细胞比例显著下降,细胞凋亡率明显增加(均P<0.05)。结论:CD44 siRNA干扰载体可高效抑制鼻咽癌SUNE-15-8F细胞CD44的表达,诱导细胞阻滞在G0/G1期,促进细胞凋亡。
Objective:To explore the effects of CD44 siRNA on cell cycle and apoptosis in nasopharyngeal carcinoma.Methods:CD44 siRNA fragment was designed,synthesized and transfected into human nasopharyngeal carcinoma SUNE-15-8F cells.The transfection efficiency was assessed by Real-time qPCR and Western blot.The cell cycle and apoptosis were detected by flow cytometry.Results:CD44 siRNA decreased the expression of CD44 mRNA and protein significantly in SUNE-15-8F nasopharyngeal carcinoma cells(P<0.05).The proportion of G0/G1 phase cells was obviously increased,while the proportion of S phase cells was greatly decreased,with significant elevation of apoptosis rate in the siRNA group,when compared with the control group(all P<0.05).Conclusion:CD44 siRNA interference carrier could effectively inhibit the expression of CD44 in nasopharyngeal carcinoma cell,which can block cell cycle in G0/G1 phase,and enhance cell apoptosis.
