摘要
分别使用四种不同的方法将替考拉宁和万古霉素键合在硅胶表面,制备了八种不同的高效液相色谱柱固定相,并在反相模式和极性有机相模式下考察了这八种手性柱的性能。实验表明,固定相制备方法的不同和色谱条件的改变对色谱柱拆分性能有一定的影响,不相同的键合臂,能拆分的对映异构体可能不一样,这些柱之间具有一定的互补性。八种手性固定相均有一定的手性拆分效果,特别是对α-氨基酸及其衍生物具有较好的手性识别能力,所制备的手性柱可用在高效液相色谱法拆分手性化合物中。
Teicoplanin and vancomycin were bonded on the surface of silica gel by four different methods to prepare eight columns for high performance liquid chromatography.Their enantioseparation properties were investigated in reversed-phase mode and polar organic phase mode.The results show that the different preparation methods of stationary phase and the change of chromatographic conditions have a certain impact on the resolution performance of chromatographic columns.The enantiomers that can be separated may be different with different bonding arms,and there is a certain complementarity between these columns.All the eight chiral stationary phases possess chiral recognition ability,especially for the enantiomers ofα-amino acids and their derivatives.The prepared chiral column can be used for the resolution of chiral compounds in high performance liquid chromatography.
